椒蒿挥发油对小鼠自身免疫性睾丸炎的影响

摘　　要：目的研究椒蒿挥发油对小鼠自身免疫性睾丸炎睾丸组织结构及血清抑制素B和卵泡刺激素含量的影响。方法健康成年雄性BALB/c小鼠150只,随机分为生理盐水组、模型组及椒蒿低、中、高剂量组(即椒蒿挥发油的体积分数为0.938%、1.875%、3.375%),采用单侧睾丸注射冰醋酸的方法造模。各组都在造模前3d开始灌胃,手术并单侧睾丸注射冰醋酸后继续灌胃,连续灌胃24d后,分别于术后3周、6周、9周取材检测,光镜观察非手术侧睾丸生精小管内的变化,ELISA法检测血清抑制素B和卵泡刺激素的变化情况。结果术后3、6、9周,模型组及椒蒿低、中剂量组血清抑制素B含量均低于生理盐水组,差异有统计学意义(P<0.05),卵泡刺激素含量均高于生理盐水组,差异有统计学意义(P<0.05),术后9周,椒蒿高剂量组血清抑制素B和卵泡刺激素含量与生理盐水组比较,差异无统计学意义(P>0.05),术后9周,椒蒿高剂量组光镜下睾丸组织结构基本正常。结论椒蒿挥发油对小鼠自身免疫性睾丸炎的发生有一定的拮抗作用,同时也跟剂量有关,高剂量组的拮抗效果最好,术后灌胃9周,睾丸组织结构基本恢复正常。ABSTRACT. Objective To study the effects of pepper Artemisia volatile oil on testicular tissue structure as well as serum inhibin B and follicle stimulating hormone （FSH） contents in experimental autoimmune orchitis in mice. Methods Totally 150 adult male BALB/c mice were randomly divided into normal saline group, model group, and low-, medium- and high-dose pepper Artemisia volatile oil groups. The volume fraction of pepper Artemisia volatile oil was 0. 938%, 1. 875% and 3. 375%, respectively. The model was established by unilateral testicular injection of glacial acetic acid method. Each group began gavage 3 days before the surgery, continued gavage after surgery and lasted 24 days. After 3, 6 and 9 weeks, changes in testicular seminiferous tubules on the un-operated side were observed under optical microscope; serum inhibin B and FSH changes were detected by ELISA. Results Serum inhibin B content was significantly lower in model group, low-dose group and medium-dose group than in saline group 3, 6 and 9 weeks after the surgery （P^0.05） ; FSH content was significantly higher than that in saline group （P〈0.05）. Nine weeks after the surgery, serum inhibin B and FSH contents in high-dose group did not differ significantly from those in saline group （P〉0.05）. Under the light microscope, the testicular tissue structure in high-dose group returned to normal. Conclusion Pepper Artemisia volatile oil has an antagonistic effect on experimental autoimmune orchitis in mice in the dose-dependent manner. Gavage for 9 weeks after the surgery makes the testicular tissue structure almost recover.
