摘 要:目的研究通脑溶栓胶囊对大鼠体内血栓形成和血液流变学的影响。方法 SD大鼠随机各分为5组:阴性对照组、阳性对照组及通脑溶栓胶囊低、中、高剂量组,阴性对照组灌胃给予5g/L的CMC-Na,其余各组灌胃等体积对应的受试品混悬液,连续给药30d,观察通脑溶栓胶囊对动、静脉血栓形成的影响;观察对血小板聚集及凝血功能的影响;另SD大鼠随机分为6组:阴性对照组、模型对照组、阳性对照组及通脑溶栓胶囊低、中、高剂量组,连续给药30d,末次给药后尾静脉注射右旋糖酐,腹主动脉取血,检测血液流变学指标。结果与阴性对照组比较,通脑溶栓中、高剂量组血栓湿重明显减轻(P<0.05或P<0.01);通脑溶栓中剂量组能延长血凝时间(P<0.05),中、高剂量组可降低血小板聚集率,延长凝血酶原时间,低、高剂量组可延长活化部分凝血活酶时间(P<0.05或P<0.01);与模型对照组比较,通脑溶栓中、高剂量组能明显降低全血中切、高切、低切及血浆黏度(P<0.05或P<0.01)。结论通脑溶栓胶囊能明显抑制血栓的形成,降低血小板聚集率及血液黏度,改善血液流变学而发挥活血化瘀作用。Objective To study the effect of Tong-nao thrombolysis capsule on thrombus formation in rats in vitro and blood viscosity in rats. Methods SD rats were randomly divided into five groups: negative control group, positive control group, as well as Tong-nao thrombolysis capsule low-, medium- and high-dose groups. Negative control group was intragastrically administered with 5 g/L CMC-Na while the rest groups were intragastrically administered with the corresponding volume of the tested suspension for thirty consecutive days. The effects of Tong-nao thrombolytic capsule on arterial and venous thrombosis, platelet aggregation and blood coagulation in rats were investigated. Other SD rats were randomly divided into six groups: negative control group, model group, positive control group, as well as Tong-nao thrombolysis capsule low-, medium- and high-dose groups. They were intragastrically administered with the drugs for one month continually. Dextran was injected intravenously via the tail vein after the final administration. Blood was taken via abdominal aorta and the hemorheological indices were detected. Results Compared with that in negative control group, the wet weight of thrombus was significantly reduced in Tong-nao thrombolysis capsule medium- and high-dose groups (P〈0.05 or P〈0.01). Blood coagulation time was prolonged in Tong-nao thrombolysis capsule medium-dose group (P〈0.05). Plasma prothrombin time was also prolonged in Tong-nao thrombolysis capsule medium- and high-dose groups (P〈0.05) In Tong-nao thrombolysis capsule low- and high-dose groups, activated partial thromboplasti^a time was prolonged (P〈O. 05 or P〈O. 01). Compared with those in model control group, the whole blood middle, high and low shear rates as well as plasma viscosity were markedly decreased in Tong-nao thrombolysis capsule medium- and high-dose groups (P〈0.O5 or P〈0.01). Goflclusion Tong-nao thrombolysis capsule can significantly inhibit the formation of blood clots, reduce platelet aggregation
