摘  要:目的:研究并建立参茶软胶囊中丹参酮Ⅱ_A和总黄酮的含量测定方法。方法:采用HPLC法测定丹参酮Ⅱ_A含量,色谱柱为Optimusil C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(75:25),流速为0.8 ml·min^(-1),检测波长为270 nm;采用紫外可见分光光度法测定总黄酮含量,以芦丁为对照品,于360 nm波长处测定。结果:丹参酮Ⅱ_A在0.2~3.6μg·ml^(-1)范围内呈良好的线性关系(r=0.999 8),芦丁在0.002 9~0.029 5 mg·ml^(-1)范围内呈现良好的线性关系(r=0.999 9);丹参酮Ⅱ_A和芦丁的加样回收率分别为98.77%、99.21%,RSD分别为0.337%、0.353%。三个批号的参茶软胶囊中丹参酮Ⅱ_A和总黄酮的平均含量分别为141.92μg·g^(-1),25.22 mg·g^(-1)。结论:本方法简便易行,准确可靠,重复性好,可以用于参茶软胶囊及其类似产品中丹参酮Ⅱ_A和总黄酮的质量控制。Objective:To establish a method to determine the content of tanshinone ⅠA and total flavonoids in Shencha soft capsules.Method:The content of tanshinone ⅡA was determined by HPLC using an Optimusil C18 column.The mobile phase was methanol-water(75:25) with a flow rate of 0.8 ml·min-1 and a temperature of 30℃.Tanshinone ⅡA was detected at wavelength of 270 nm.The content of total flavonoids in Shencha soft capsules was determined by UV-visible spectrophotometry at 360 nm.Rutin was used as the reference substance.Result:The average content of tanshinone ⅡA and total flavonoids in three batches of Shencha soft capsules was 141.92 μg?g-1 and 25.22 mg?g-1,respectively.The linear equation of tanshinone ⅡA was Y = 4.35×104X+3.06× 102 within the range of 0.2-3.6 μg?ml-1(r = 0.999 8).The linear equation of rutin was Y=19.1X-4.4×10-3 within the range of 0.002 95-0.029 5 mg ? ml-1(r = 0.999 9).The average recovery and RSD of tanshinone ⅡA and rutin were 98.77%and 99.21%, 0.337%and 0.353%,respectively.Conclusion:The established method is simple,accurate and reliable with good reproducibility.It can be used lo control the quality of tanshinone ⅡA and total flavonoids in Shencha soft capsules and other similar products.
