摘 要:目的探讨八味沉香散对大鼠心肌缺血/再灌注损伤后心肌细胞凋亡相关基因表达的影响。方法采用左冠状动脉穿线结扎法制备心肌缺血/再灌注模型。SD大鼠40只随机分成4组:假手术组(A组,n=10),缺血/再灌注损伤模型组(B组,n=10),缺血/再灌注损伤模型+生理盐水对照组(C组,n=10),八味沉香散组(D组,n=10)。心电图记录肢体Ⅱ导联ST段的变化。利用RT-qPCR检测心肌Bcl-2、Bax基因的mRNA表达情况。结果缺血/再灌注损伤模型组与假手术组比较,Bcl-2下降,Bax增高,Bcl-2/Bax下降(P<0.05);缺血/再灌注损伤模型+生理盐水对照组各项指标与缺血/再灌注损伤模型组比较均无显著性差异;八味沉香散组的各项指标较缺血/再灌注损伤模型组明显改善,Bcl-2升高,Bax下降,Bcl-2/Bax升高。结论八味沉香散可能通过上调Bcl-2、下调Bax来抑制缺血/再灌注后心肌细胞凋亡,从而对缺血/再灌注心肌损伤发挥保护作用。Objective To explore the possible protective mechanism in vivo of Tibetan herb Ba Wei Chen Xiang in Myocardial tissue of myocardial ischemia and reperfusion injury rats. Methods 40 SD rats were randomly divided into sham operation ( group A, n = 10), myocardial ischemia & reperfusion (MIR) group ( group B, n = 10 ), MIR + physiological saline group ( group C, n = 10), MIR + Ba Wei Chert Xiang group ( group D, n = 10). ST seg- ment changes in lead II were recorded. The expression of apoptosis related genes Bcl - 2 and Bax were detected with the methods of RT - qPCR. Results Bax increased while Bcl - 2 decreased, the ratio of Bcl - 2/Bax decreased sig- nificantly in group B compared with that in group A ( P 〈 0.05 ). There were no significant difference between group B and group C. Compared with group B, Bax decreased while Bcl -2 increased and the ratio of Bcl -2/Bax in- creased in group D (P 〈 0.05 ). Conclusion Tibetan herb Ba Wei Chen Xiang is likely to protecting the ischemia/reperfusion myocardial injury by up -regulating Bcl- 2 and down- regulating Bax to inhibit ischemia/reperfusion myocardial apoptosis.
