摘 要:目的分析细粒棘球绦虫幼虫生长环境即囊液的蛋白质构成情况,为深入了解棘球蚴的发育、代谢奠定基础。方法采用双向凝胶电泳分离蛋白质,经过图像分析标记,通过自动切胶仪切胶、回收,胶内酶解、纯化以及质谱分析技术分析蛋白样点的数据,最后结合生物信息学方法进行注解。结果在囊液二维电泳图谱上提取约30个蛋白斑点,成功获取21种蛋白的肽指纹图谱,利用生物信息学检索工具MASCOT进行了蛋白鉴定。结论通过建立细粒棘球蚴囊液的二维电泳图谱,初步鉴定出囊液中含人血清铁传递蛋白、人白蛋白、人β-血红蛋白3种蛋白,为从外界干扰囊液中蛋白质构成比方面寻找包虫病治疗的新方法提供了参考依据。Objective To analyze the protein component of hydatid cyst fluid of echinococcus granulosus so as to establish the base for development and metabolism of echinococcus. Methods The protein was separated with two-dimensional gel electrophoresis method,marked with image analysis; gel was cut and the dots were collected with automatic machine by choosing the target protein,and the data of protein dot was analyzed by matrix-assisted laser desorption ionization time of flight mass spectrometry( MALDI-TOF-MS). Every protein was identified to be combined with the database record of peptide mass fingerprint( PMF) by Mascot search engine. Results A total of 30 protein spots were extracted from two-dimensional electrophoretogram of the hydatid fluid,and 21 peptide fingerprints of protein were successfully obtained. The protein was identified by using of MASCOT. Conclusion Serum transferrin,human albumin and human β- hemoglobin in the hydatid fluid can be preliminarily identified by establishing two-dimensional electrophoresis mass spectrometry of hydatid cyst fluid. It may provide theory evidence for new ways to treat hydatid disease by inhibitting or blocking the protein constituent ratio in hydatid fluids.
