摘 要:目的探讨苯并芘氧化损伤肺泡细胞的作用机制及黄芪注射液是否对其有保护作用。方法选择性成熟雄性SD大鼠100只,随机分为A、B、C、D、E组5组,每组20只,前4组予苯并芘1 mg/(kg·d)灌胃,B、C、D组灌胃苯并芘同时静脉注射10、20、40 g/(kg·d)黄芪注射液,E组灌胃橄榄油并静脉注射0.9%氯化钠注射液。连续静脉注射15 d后处死大鼠,取肺泡组织测定氧化和抗氧化物质。结果 B、C、D组肺泡组织中谷胱甘肽过氧化物酶(glutathione peroxidase,GSHPX)、超氧化物歧化酶(superioxide dismutase,SOD)活性明显高于A组,但低于E组,差异均有统计学意义(P<0.05);B、C、D组肺泡组织活性氧类物质(reactive oxygen species,ROS)活性及丙二醛(malondialdehyde,MDA)含量低于A组,高于E组,差异均有统计学意义(P<0.05)。结论黄芪注射液能抑制苯并芘所致的肺泡组织GSH-PX、SOD活性降低及ROS活性、MDA含量的升高。Objective To explore the mechanism of action for oxidative damnification of alveolar tissues induced by BaP and investigate whether astragalus membranaceus plays a protective role or not.Methods Viripotent 100 male SD rats were randomly divided into group A,B,C,D and E (n =20).The rats were lavaged with 1 mg/(kg · d) BaP in group A,B,C and D,while the rats in group A,B and C were given additional astragalus injection with a dose of 10,20 and 40 g/(kg · d) respectively,and group E was lavaged with olive oil and injected with 0.9% sodium chloride injection.The oxydic and antioxidative indexes were detected after continuous intravenous injection for 15 d.Results The activities of glutathione peroxidase (GSH-PX) and superoxide dismutase (SOD) in alveolar tissues in group B,C and D were significantly superior than those in group A,but were significantly lower than those in group E (P < 0.05); the activities of reactive oxygen species (ROS) and the concentration of malondialdehyde (MDA) in alveolar tissues in group B,C and D were significantly inferior to those in group A,but were significantly superior to those in group E (P < 0.05).Conclusion Astragalus membranaceus can inhibit the depressed activity of GSH-PX and SOD induced by BaP,and can inhibit the raised activity of ROS and concentration of MDA induced by BaP in alveolar tissues.
