摘  要:目的观察中药组方糖脉通对糖尿病模型大鼠血管组织肥胖抑制素(Obestatin)mRNA表达的影响并探讨其机制。方法将60只雄性SD大鼠用链脲佐菌素(STZ)联合高脂饮食制造糖尿病模型大鼠,选取40只随机分为模型对照组、西药对照组、糖脉通小剂量组、糖脉通大剂量组4组,每组各10只;另取10只正常大鼠为空白对照组。模型对照组以0.9%氯化钠注射液10 mL/kg灌胃给药;糖脉通小剂量组在格列齐特20 mg/kg的基础上,以生药浓度为2 g/mL糖脉通水煎剂10 mL/kg灌胃给药;糖脉通大剂量组在格列齐特20 mg/kg的基础上,以生药浓度为4 g/mL糖脉通水煎剂10 mL/kg灌胃给药;西药对照组在格列齐特20 mg/kg的基础上,以浓度为1.8 mg/mL的西洛他唑混悬液10 mL/kg灌胃给药;空白对照组以等容积蒸馏水灌胃。除空白对照组外均给予高糖高脂饲料。药物干预12周后,采用逆转录-聚合酶链反应(RT-PCR)方法检测各组大鼠血管组织Obestatin mRNA的表达。结果模型对照组、西药对照组及糖脉通小剂量组大鼠血管组织中Obestatin mRNA表达水平较空白对照组明显升高(P<0.05);糖脉通大、小剂量组及西药对照组血管组织中Obestatin mRNA表达较模型对照组减少(P<0.05),且糖脉通大、小剂量组效果优于西药对照组(P<0.05)。结论糖脉通可能通过降低Obestatin mRNA在模型大鼠血管组织中的异常表达,对血管功能起到保护作用。Objective To observe the effect and mechanisms of Tangmaitong on mRNA expression of Obestatin in the vascular tissue of diabetic rat' s model. Method Rat diabetic model were made with STZ and high - quality and high carbohydrate diet. 40 diabetic model rats were randomly divided into four groups, model group, western medicine group, high- and low- dose Tangmaitong group. 10 normal rats were as control group. Model group received 0.9% NaC1 solution,high- and low- dose Tangmaitong groups received 10mL/kg(2 g/mL,4 g/mL crude drug) on the basis of Gliclazide,20mg. kg. Rat in western medicine group received 10 mL/kg( Cilostazol suspension 1.8 mg/mL) on the basis of Gliclazide,20m~. k~. Control zrouo received eaual volume of distilled water. After 12weeks of drug intervention, rats were killed, the vascular were removed and the mRNA expression of Obestatin in the vascular was observed by the method of RT - PCR. Results The mRNA expression of Obestatin was signifi cantly increased, and Tangmai -tong could lower the mR NA expression of Obestatin in the vascular in a dose - de pendent manner. The decrease of the mRNA expression of Obestatin in high - and low - dose Tangmaitong group was superior to that in western medicine group ( P 〈 0.05 ). Conclusion Tangmaitong can play a protective effect onvascular function by reducing the abnormal expression of Obestatin in the vascular tissue of diabetic rat" s model, and deserves further research.
