摘  要:为深入研究福建省地方鸭品种的群体遗传结构,本研究利用经过筛选验证的荧光标记微卫星引物结合ABI-3730XL型DNA测序平台,对连城白鸭、莆田黑鸭、金定鸭、山麻鸭和福建番鸭等5个品种共计464个个体在12个位点上的等位基因进行STR分型,分别利用SPSS 19.0和Structure2.0软件对群体结构进行分析。结果显示:12个座位的多态性丰富,共检测到81个等位基因,PCA分析得到4个主成分,前3个主成分对群体分类的累积贡献率达到85.904%。Structure程序成功构建出了5个群体的遗传分化结构图和群体的独立类群分布图,推断5个群体在所属推断类别中的平均基因组分数分别为94.7%、95.9%、92.4%、93.3%、97.8%;且只有4个个体发生了迁移。Structure程序构建的系统发育树显示4个河鸭属品种遗传距离较近,与福建番鸭亲缘关系较远;山麻鸭与金定鸭、莆田黑鸭与连城白鸭各自聚类后,再与福建番鸭聚类。In this study, the allele frequency of 12 microsatellite loci in Liancheng, Putian, Jinding, shanma, and Fujian muscovy duck were detected by STR genotyping method based on ABI-3730XL DNA Sequencing platform. The quantity of all individuals was 464. The alleles were analyzed by SPSS 19.0 and Structure2.0 software. The results showed that the polymorphisms of all the 12 loci were abundant and 81 alleles were obtained. The distribution of the alleles was analyzed by principal component analysis and 4 principal components were got. The accumulating contribution rate of the first 3 principal components was 85.904%. The genetic differentiation structure and distribution of independent groups of 5 indigenous populations were structured by structure procedure. The average gene components in their supposed groups were 94.7% ,95.9% ,92.4% ,93.3% ,97.8%, and only 4 individuals were migrated into another population. The phylogenetic tree showed that the genetic distances between the populations were nearer than that between 4 populations and Fujian muscovy duck. Shanma and Jinding duck,Putian and Liancheng duck were respective clustered firstly and then clustered with Fujian muscovy duck.
