摘 要:为研究骨保护素(Osteoprogerin,OPG)对体外培养鸭胚破骨细胞(Osteoclast,OC)分化与活化的影响,收集23日胚龄鸭胚骨髓细胞,体外培养过程中分组加入0 ng/mL(A组)、30 ng/mL(B组)、100 ng/mL(C组)OPG进行处理。通过抗酒石酸酸性磷酸酶染色(Tartrate-resistant acid phosphatase,TRAP),骨吸收陷窝分析技术,检测TRAP阳性细胞数量及OC的骨吸收活性。结果显示,3个试验组TRAP阳性细胞数、OC的骨吸收活性均在OPG处理7 d后达到最高水平。与A组相比,OPG处理组OC数目及其骨吸收活性均显著减弱(P<0.05),每个时间段骨吸收陷窝净增长面积与OC数目变化趋势一致。表明OPG能够抑制OC细胞前体分化,促进成熟OC发生凋亡,抑制成熟OC的骨吸收活性。To determine the influence of osteoprotegcrin (OPG)on differentiation and activation of duck embryo osteoclasts cultured in vitro, bone marrow cells were harvested from 23-day-age duck embryos and cultured in the presence of different concentrations of OPG,such as no added factors (group A),30 ng/mL OPG (group B)and 100 ng/mL OPG (group C). Tartrate-resistant acid phosphatase (TRAP)staining and bone resorption activity assay were performed to determine the number of TRAP-positive cells and bone resorption activity of duck embryo osteoclasts. The results showed that the number of TRAP-positive cells and the net expansion of pit formations area peaked were on the 7th day of culture in all three groups. The number of osteoclasts and the bone resorption activity of them in OPG-treated groups were significantly lower compared to group A (P〈0.05). At each time point,the net expansion of bone resorption area was correlated with the number of TRAP-positive cells. The results indicated that OPG inhibited the differentiation of osteoclast precursors, and in mature osteoclasts OPG enhanced the development of apoptosis, decreased the bone resorption activity.
