摘 要:目的:建立乌头碱的液相色谱-质谱联用(LC-MS-MS)含量测定方法,测定乌头碱微透析体内回收率并考察其稳定性。方法:采用反透析法测定回收率,考察流速、浓度、时间对回收率的影响。结果:反透析法测定的回收率与浓度、时间无关,稳定性良好,随流速增加回收率降低。结论:微透析取样技术可用于乌头碱的药动学研究,体内研究的反透析法可作为微透析研究中乌头碱回收率的测定方法。Objective: To establish an LC-MS-MS method to determine the in vivo recovery of aconitine in microdialysis probes and study its stability. Method: The recovery of aconitine was detected by retrodialysis method. The effect of flow rates, concentrations and time on recovery was studied. Result: At the same concentration, the recovery decreased with the increase of the flow rate. Aconitine recovery was independent of aconitine concentration in the external medium by using retrodialysis method. Aconitine recovery had good stability. Conclusion: Microdialysis sampling can be used for the pharmacokinetic study of aconitine and retrodialysis method can be used for the determination of aconitine recovery in vivo.
