摘  要:利用响应面分析方法对铁皮石斛组培苗多糖的提取工艺进行优化。在单因素试验的基础上选取试验因素与水平,根据中心组合(Box-Behnken)试验设计原理采用三因素三水平的响应面分析法,确定各工艺条件的影响因素,以铁皮石斛组培苗多糖的提取率为响应面和等高线,在分析各个因素的显著性和交互作用后,得出铁皮石斛多糖浸提的最佳工艺条件为:温度92℃、料液比1∶41、浸提时间1.6 h,在最佳提取条件下,浸提3次,铁皮石斛的多糖得率为38.569%。Based on single factor experiments, a three-variable, three-level Box-Behnken experimental design and re- sponse surface methodology were applied to optimize the extraction process of water-soluble polysaceharides from tissue culture seedling of Dendrobium officinale. Response surface and contour plots were obtained using the extraction yield of polysaccharide as indicator. In addition, the analysis of significance of process conditions on polysaceharide extraction and their interactions was carried out. The optimal extraction conditions were determined to be:extraction temperature of 92 ℃, solid/liquid ratio of 1:41 and with 1.6 h of extraction. Under the optimized extraction conditions, the extraction yield of polysaccharide was 38. 569%.
