摘  要:目的:为筛选二苯乙烯苷含量差异显著的何首乌样本,为通过mRNA水平筛选与二苯乙烯苷合成相关酶基因,研究二苯乙烯苷合成途径打下基础。方法:HPLC,流动相为乙腈∶水(25∶75);柱温:27℃;流速:1.0mL.min-1;进样量:10μL;紫外检测波长:320nm;进样时间:10min。结果:线性回归方程:y=41.233x+1.6644,R2=1。结论:对水浴和室温冷浸两种萃取方法的比较发现水浴40min和冷浸12h效果无明显差异;冷浸12h和冷浸30h无显著差异;不同产地何首乌块根二苯乙烯苷含量差异显著(6.4%~0.0011%),同一产地何首乌含量差异亦存在明显差异(6.4%~1.5%);同株何首乌不同组织二苯乙烯苷含量存在较大差异,依次是:根〉茎〉叶;新叶〉老叶;老藤〉新藤。Objective:To screen samples the difference of 2,3,4,5' -tetra- hydroxy- stilbene- 2- O- β -D- glu- coside content for screening the genes correlated with the synthesis of 2,3,4,51 -tetra -hydroxy -stilbene -2 -O -β - D- glucoside at mRNA level and to research the pathway of 2,3,4,51 -tetrahydroxy- stilbene- 2 -O-β-D -glueoside synthesis. Methods.HPLC was used to determine the content. The mobile phase was H2O and CH3 CN with the ratio of 3 - 1 (v/v) over 10 rain. The flow rate was 1 mL/min, with detection at 320 nm. Results. The linear equation was : y = 41. 233x + 1. 6644, R2 = 1. Conclusion :The comparison of water bath for 40 rain and quench at room temperature for 12 h shows that there is no obvious difference. There is no distinct difference between quench at room temperature for 12 h and 30 h either. The content is distinctly different in root tuber from different areas, and even different in root tubers from the same area. The content in different tissues is : root 〉 stem 〉 leaf; young leaf 〉 old leaf; old stem 〉 young stem.
