摘 要:目的:检测肝组织中TT病毒,并探讨其致病性.方法:采用对称和不对称PCR法,以地高辛素分别标记TTV双链和单链探针,原位杂交法检测56例肝组织。结果:51例非甲~非庚型肝炎肝组织中TTV总检出率27.5%(14/51);5例非肝炎肝组织杂交阴性。TTV DNA主要见于肝细胞核内,在急性、慢性及重型肝炎组织中均有检出。双链探针杂交结果与单链探针检测病毒基因链的结果一致,2/6例组织TTV基因链与互补链同时存在.结论:TTV是导致肝炎的一种新病原,井且在肝脏内有复制。Aim:To study Transfusion Transmitted Virus(TTV) infection from liver tissues. Methods:By symmetrical and unsymmetrical polymerase chain reaction (PCR), double-strand and single-strand TTV probes were labelled with Digoxigenin. 56 liver samples were detected by in situ hybridization. Results: 14 of 51 patients with non-hepatitis A -non-hepatitis G were found TTV DNA (27- 5%); Five liver samples without hepatitis were negative. TTV DNA was mainly observed in nuclei of hepatocytes and distributed in acute, chronic and severe types of hepatitis. The hybridization result by double-strand probe was consistent with TTV gencnie strand hybridization by single-strand probe. In 2 of 6 liver samples were present TTV genome strand and complementary strand. Conclusion: This study indicates that TTV is a noval virus which resulting in hepatitis; The virus replicates in liver tissue.
