医用石蜡油在荧光PCR检测HBV—DNA定量质量控制中的应用

摘　　要：目的探讨无菌医用石蜡油在荧光PCR检测HBVDNA定量质量控制中的应用价值。方法经荧光定量PCR扩增好的江苏省临检中心的质控物高低值为室内质控及患者的标本分成组，及将患者的提取物分为两份，一份加定量浓度的无菌石蜡油封管与另一份不封管，以荧光定量PCR技术扩增原理，采用分组测定HBV—DNA的含量，连续测定6个月实验，根据CT值统计每组的SD，CV，P值的情况。结果江苏省临检中心提取好的质控物经无菌石蜡油分管扩增的产物测定特别稳定，加石蜡油组CV为2．92％，SD为0．126而不加组CV为7．12％，SD为0．333，两组差异有统计学意义。结论无菌医用石蜡在PCR质控的保存封管扩增能起到结果稳定，提高了PCR测定结果的质量，有利于PCR测定的技术的普及和推广应用。Objective To explore the application value of sterile medical paraffin oil in the quality control, of HBV DNA fluorescent quantitative PCR analysisMethods By fluorescence quantitative PCR amplification of clinical inspection center of Jiangsu province quality control substance of value of indoor air quality and the patients were divided into two groups, and the patient＇s extract is divided into two parts, a seal paraffin and quantitative concentration tube and another not sealed tube amplification principle,using fluorescence quantitative PCR technology, content determination of HBV-DNA by grouping,continuous determination of 6 months experiment,according to the statistics of CT value with SD,CV,P value. Results Quality control of Jiangsu province clinical inspection center extraction by product aseptic paraffin oil in the amplification of the determination of particularly stable,and paraf~.in oil groups＇ CV was 2.92~/oo ,SD was 0. 126,comparing with the no oil groups＇ CV was 7.12~ ,SD was 0. 333,there was a significant difference between the two groups. Conclusion The medical sterile paraffin is added in the PCR control of the sealed tube amplification can play a stable result, improve the examnation of PCR quality, popular for PCR measurement technology and application.
