摘 要:目的探讨长期应用大黄及其总蒽醌对雄性未成年大鼠生殖功能的影响。方法 3周龄SD大鼠50只,随机分为空白对照组,大黄水提物小、中、大剂量组(0.3、0.6、1.2g/kg)及大黄总蒽醌组(0.1g/kg)。各组连续灌胃给药30天,取睾丸计算睾丸指数并进行病理学检查,检测精子数量、活动率及畸变率。采用ELISA法检测血清睾酮(T)、黄体生成素(LH)、卵泡刺激素(FSH);采用免疫组化SP法检测睾丸bcl-2、bax蛋白表达水平的变化。结果与空白对照组比较,大黄水提物中、大剂量组和大黄总蒽醌组睾丸、附睾指数降低;各给药组大鼠精子数量和活动率均降低;血清中T水平降低,LH及FSH水平升高;睾丸生精上皮和睾丸间质细胞内bax蛋白表达升高,而bcl-2蛋白表达降低;差异均有统计学意义(P<0.05)。大黄水提物各剂量组的精子数量、活动率,血清性激素T、LH、FSH水平及bax、bcl-2表达比较,差异均有统计学意义(P<0.05)。大黄水提物各剂量组和大黄总蒽醌组大鼠睾丸病理组织学可观察到间质细胞、生精上皮细胞的破坏。结论长期应用大黄对未成年大鼠生殖功能有较强的毒性,且毒性反应程度有明显的剂量依赖关系。Objective To discuss the clinical efficacy of Rheum and total anthraqainone on reproductive function of minor male rats. Methods Fifty SD rats aged three weeks were randomly divided into blank control group, Rheum extracts groups (0.3, 0.6, 1.2 g/kg) and total anthraquinone group (0.1 g/kg). The rats were given rheum extracts by gastric administration for 30 days continuously. Then taken out testes and epididymis to detect the sperm count, rates of sperm mobility and aberration. The levels of T, LH and FSH in serum and the expressions of bcl-2 and bax in testes were measured by ELISA and immunohistochemistry SP method, respectively. Results Compared with blank control group, the indexes of testis and epididymis in medium dose, high dose of Rheum extracts groups and total anthraquinone group were decreased ; the sperm count and rate of sperm mobility in Rheum extracts groups and total anthraquinone group were decreased ; the T level was decreased and levels of LH and FSH were increased ; the expression of bax in seminiferous epithelium and leydig cells were increased, while the bcl-2 expression was decreased significantly ( P 〈 0.05 ). There were significant differences in amount of sperms, activity ratio and levels of T, LH, FSH, bax expression, bcl-2 expression among Rheum extracts groups (P 〈 0. 05 ). Histopathologically, the damage of mesenehymal cells and seminiferous epithelial cells can be observed in the testes of the Rheum extracts groups and total anthraquinone group. Conclusion Rheum has toxic reaction to minor rat testes with significant dose-dependent effect.
