摘  要:目的研究在Fas通路中参与调控诱导EMT的相关分子在结直肠癌临床样本中的表达水平及其与肿瘤的临床病理指标、肿瘤预后之间的关系,并以此为基础探索结直肠癌中Fas诱导EMT的可能机制。方法采用免疫印迹、免疫组化技术检测结直肠癌患者组织标本中FasL、p-GSK3β、Snail和β-catenin的表达水平,并统计其表达水平与患者临床病理指标之间的关系。将临床标本根据这四种分子表达水平的高低分为EMT表型和非EMT表型,统计分析该分型与肿瘤分期及患者预后之间是否相关。分析p-GSK3β、Snail和β-catenin之间的两两相关性,以探索结直肠癌中Fas诱导EMT的可能机制。结果对人结直肠癌标本进行评分后发现,FasL、p-GSK3β、Snail和β-catenin的表达水平与肿瘤分期呈正相关。与非EMT表型相比,EMT表型的患者肿瘤分期较高且预后较差。相关性分析显示,p-GSK3β、Snail和β-catenin在结直肠癌组织中的表达水平两两相关。结论 p-GSK3β、Snail和β-catenin可能参与调控Fas诱导的EMT过程,从而促进结直肠癌转移。Objective To explore the clinical significance of Fas-induced-EMT associated molecules in colorectal cancer, and to investigate the possible molecular mechanisms of Fas-induced EMT in colorectal cancers. Methods Western blot and immunohistochemistry were used to detect the expression levels of FasL, p-GSK3β, Snail and β-catenin in clinical samples of patients with colorectal cancer. The association between the expression levels of the four molecules and the clinicopathologic parameters of colorectal cancer was ana- lyzed. We defined the clinical specimens as EMT phenotype or non-EMT phenotype according to the expres- sion levels of these four molecules, and analyzed the association between the phenotype and cancer stage or prognosis. Spearman correlation was used to analyze the pair-wise correlation among p-GSK3β, Snail and β- catenin. Results FasL, p-GSK3β, Snail and β-catenin expression levels were increased along with cancer progression. EMT phenotype was related to advanced tumor staging and poor prognosis comparing with non- EMT phenotype. Spearman analysis showed significant correlation between p-GSK3β, Snail and β-catenin. Conclusion p-GSK3β, Snail and β-catenin may be involved in the regulation of Fas-induced EMT process, which promote colorectal cancer metastasis.
