摘  要:肾小球系膜细胞(mesangial cells,MCs)增殖、肥大在糖尿病肾病(diabetic nephropathy,DN)的发展中处于重要地位。胰岛素可通过胰岛素信号途径促进MCs增殖,增强MCs分泌细胞外基质(extracellular matrix,ECM)、促进ECM积聚,从而在DN发生发展过程中处于十分重要的地位。MCs中存在Wnt信号通路,以维持MCs的正常生理功能。异常的Wnt通路导致MCs凋亡过多或肾纤维化。胰岛素和Wnt信号通路存在相互作用。越来越多的证据表明,高胰岛素环境中MCs内存在Wnt和胰岛素信号通路的相互作用,调控MCs增殖从而影响DN发生发展。深入研究Wnt信号通路和胰岛素信号发生相互作用,并对MCs增殖的分子机制产生影响开发新药物,通过调控Wnt信号通路改善肾小球MCs增殖,将为2型糖尿病肾病的临床治疗提供新的科学依据,寻找新的治疗靶点。Proliferation and hypertrophy of glomerular mesangial cells (MCs) play an important role in the development of the diabetic nephropathy (DN). Insulin promotes MCs proliferation, enhances MCs secreting ECM (extracellular matrix), promotes ECM accumulation through the insulin signaling pathways, which is very important for the development and progression of DN. The Wnt sig- naling pathway exist in the MCs, in order to maintain the normal physiological functions of MCs. Abnormal Wnt pathway leads to exces- sive MCs apoptosis or renal fibrosis. There is a dialogue exist in insulin and Wnt signaling pathway. More and more evidences indicate that there is a dialogue existing in high insulin environment, which could affect the development of DN by the regulation of MCs prolif- eration. It will provide new scientific basis to develop new therapeutic targets for the treatment of type 2 diabetic nephropathy. Wnt signaling pathway and the dialogue and insulin signaling occurs effecting the molecular mechanism of MCs proliferation, research and development of new drugs, which improves glomerular MCs proliferation through the regnlation of the Wnt signaling pathway.
