摘　　要：目的:探讨血小板内皮细胞黏附因子-1(platelet-endothelial cell adhesion molecule-1,PECAM-1)基因小分子RNA干扰(small interfering RNA,siRNA)对人鼻咽癌辐射致耐药CNE1/R细胞增殖及凋亡的影响。方法:设计干扰RNA编码DNA特异性序列,合成siRNA片段,转染CNE1/R细胞,G418筛选出稳定转染细胞株。蛋白质印迹法检测PECAM-1 siRNA干扰后,细胞中PECAM-1蛋白表达情况;MTT法检测细胞增殖及流式细胞术检测细胞凋亡。结果:筛选出稳定转染细胞株,实验组PECAM-1蛋白表达较阴性组下调3.4倍,证实干扰有效。与阴性组及空白组相比,实验组细胞增殖能力下降,凋亡率升高。结论:PECAM-1基因siRNA可抑制CNE1/R细胞增殖,促进其凋亡。PECAM-1可能在鼻咽癌细胞辐射后生长凋亡中起作用。Objective： To investigate the effect of silencing platelet-endothelial cell adhesion molecule-1（PECAM-1） by small interfering RNA（siRNA） on cell proliferation and apoptosis of irradiationinduced drug resistance CNE1 / R cells of human nasopharyngeal carcinoma.Methods： The CNE1 / Rspecified siRNA was designed and synthesized.The purity of the transfected cells were screened out by G418.The expression of PECAM-1 was detected by western blotting.Cell growth and apotosis was assayed by MTT and flow cytometry,respectively.Results： The expression of PECAM-1 protein in CNE1 / R cells was significantly downregulated.Compared with negative group and control group,experiment group showed significantly decreased proliferation ability in CNE1 / R cells,whereas cell apoptosis rate was increased.Conclusion： PECAM-1 siRNA could inhitit the growth and promote the apoptosis of CNE1 / R cell,which suggested PECAM-1 might play a key role in CNE1 / R cell growth and apoptosis.
