摘 要:目的探讨凉血散血方对免疫性血小板减少症(Immune thrombocytopenia,ITP)模型大鼠白细胞抗原分化群40分子配体(cluster of differentiation 40 ligand,CD40L)表达变化与作用机制。方法将大鼠随机分为空白组、模型组、强的松组、凉血散血方高、中、低剂量组,用腹腔注射兔抗大鼠血小板血清(APS)法造ITP模型,用酶联免疫吸附法(ELISA)测定大鼠外周血CD40L的变化,造模前后及给药后检测ITP模型大鼠血小板数值变化。结果模型组CD40L水平明显高于空白组,血小板计数明显低于空白组,两组相比较差异有统计学意义(P<0.01);与模型组比较,各治疗组CD40L水平降低(P<0.01和P<0.05),血小板数值升高(P<0.01);中药高剂量组与强的松组比较差异无统计学意义(P>0.05)。结论凉血散血方能使ITP模型大鼠外周血小板破坏减少,其作用机制可能与下调CD40L水平有关。Objective To investigate the effects and mechanism of Liangxue Sanxue Formula on the changes of cluster of differentiation 40 ligand (CD40L) of leukocyte antigen of peripheral blood in immune thrombocytopenia (ITP) rat model. Methods The rats were randomly divided into blank group, model group, prednisone group, Liangxue Sanxue Formula high, medium and low dose group. ITP rat model was made by intraperitoneal injection with rabbit anti-rat platelet serum (APS). The changes of CD40L in peripheral blood were determined by enzyme linked im- munosorbent assay (ELISA), the platelet number was detected before and after modeled, and after treatment. Results Compared with blank group, the level of CD40L was significantly higher while the platelet number was significantly lower in model group. There were statistical significances in these indices between the two groups (P〈0.01). Compared with the model group, the levels of CD40L were decreased (P〈0.01 or P〈0.05), and the platelet numbers were increased in treat- ment groups (P〈0.01). There were no statistical significances between Liangxue Sanxue Formu- la medium dose group and the prednisone group(P〉0.05) reduce peripheral platelet destruction in ITP rats, which CD40L levels.
