摘 要:目的自制葡聚糖凝胶微型柱,建立分离测定新藤黄酸(gambogenic acid,GNA)-聚乳酸-羟基乙酸共聚物(poly lactic-co-glycolic acid,PLGA)纳米微球包封率的方法。方法采用自制葡聚糖凝胶微型柱分离PLGA纳米微球与游离药物,用浊度法判断葡聚糖凝胶微型柱对PLGA纳米微球的吸附程度;用高效液相色谱法测定PLGA纳米微球中GNA含量,并计算包封率。结果自制葡聚糖微型柱可以实现游离药物与PLGA纳米微球的分离;所测3批PLGA纳米微球的包封率平均值为82.42%。结论自制葡聚糖凝胶微型柱法简便,结果较精准,适合于GNA-PLGA纳米微球包封率的测定。Objective To establish a method for determining the encapsulation efficiency of gambogenic acid (GNA)-poly (lactic-co-glycolic acid) (PLGA) nanospheres using a self-assembled Sephadex mini col- umn. Methods Free drug was separated from PLGA nanospheres using a self-assembled Sephadex mini column, and then the adsorption of PLGA nanospheres on the Sephadex mini-column was evaluated by nephelometric analysis; the content of GNA in PLGA nanospheres was determined by high-performance liquid chromatography, and the encapsulation efficiency was calculated. Results The self assembled Sephadex mini-column enabled the separation of free drug from PLGA nanospheres; the mean encapsula- tion efficiency of three samples of PLGA nanospheres was 82.42%. Conclusion The established method is simple and accurate and can be used for determining the encapsulation efficiency of GNA-PLGA nanospheres.
