摘 要:设计了一种基于磁性微球与核酸适体的夹心式化学发光适体传感器,建立了高灵敏度的可卡因分析方法。实验考察了反应所用羧基磁性微球、捕获探针、可卡因适体、生物素标记的报告序列以及链霉亲合素修饰的辣根过氧化物酶用量对化学发光信号的影响。优化条件下,在1.0×10^-8~1.0×10^-4mol/L范围内,化学发光信号与可卡因浓度的对数呈线性相关(r2=0.989 7),检出限为3.2×10-9mol/L。考察了共存物质中适体对可卡因的特异性识别能力,方法显示了较好的选择性。A sandwich aptamer-based chemiluminescence( CL) biosensor for sensitive determination of cocaine was developed based on magnetic microspheres as a special biomolecule immobilizing carrier. The experimental parameters,including amounts of magnetic microspheres,capture DNA,cocaine-binding aptamer,reporter DNA probes and dilution coefficient of SA- HRP were optimized. Under the optimized conditions,a good linear relationship was obtained between CL intensity and logarithms of cocaine concentration from 1. 0 × 10- 8mol / L to 1. 0 × 10- 4mol / L( r2= 0. 989 7). The detection limit was 3. 2 × 10- 9mol / L. To assess the CL aptasensors' ability to detect cocaine in complex samples,the specific recognition ability of cocaine aptasensors in the presence of human blood serum was also tested. The method showed a good selectivity.
