摘 要:以 [4x(亚洲棉×异常棉 ) ]×陆地棉的杂交后代培育的 5个种质系和 3个亲本为材料 ,用 2 2个随机引物进行了 RAPD分析。结果表明 :3个棉种具有明显的多态性差异。5个种质系均检测到来自不同棉种的 DNA特异片断 ;3个棉种间的遗传相似系数在 50 %以下。陆地棉亲本与种质系间和种质系与种质系间的相似系数在 0 .71 0~ 0 .933之间 ,平均为 0 .80 8。这些差异与回交亲本、单株选择的定向培育有关。研究结果对棉属远缘杂交育种具有一定的参考价值。RAPD (random amplified polymorphic DNA) markers generated by 22 random decamer primers were used to evaluate polymorphism and genetic similarity of five germplasm lines derived from 〔4x(G.arboreum×G.anomalum)〕×G.hirsutum and their parents. The result showed that there was abundant DNA polymorphism in three parents. Special DNA fragment of parents appeared in five germplasm lines. The similarity coefficients between different Gossypium species were lower than 50%, which were related to their center origin and genome difference. The similarity of germplasm lines with upland cotton and germplasm line ranged from 0.710~0.933.The high similarity is owing to backcrossing by G.hirsutum generation to generation.Meanwhile,individual plant selection with different breeding aims made the five germplasm lines show difference to some extent.
