摘  要:以黄瓜 (CucumissativusL .)栽培品种津研四号为受体 ,以质粒 pBI12 1为供体进行授粉后外源DNA直接导入技术的研究 ,建立起高效的黄瓜子房注射转化系统 ,获得了不同的转基因植株。试验表明 ,子房注射法的坐果率和结种率最低 ,但其转化率最高 ;黄瓜授粉后 12h注入外源基因的转化率最高 ;外源DNA溶液的浓度、pH值及DNA溶液注射体积是影响黄瓜坐果结实及基因转化率的重要因素。GUS组织化学染色结果以及Southernblot印迹杂交表明 。Cucumber (Cucumis sativus L.) cultivars Jinyansihao being chosen as the recipient and the plasmid pBI121 being chosen as donors,the research of the technique of direct exogenous DNA introduction after artificial pollination have been conducted.The genetic transformation system of cucumber with high efficiency by injecting exogenous DNA into ovary after pollination were established,and transgenic plants were obtained.Studies showed that the fruit setting ratio and the number of seed per fruit were the lowest by the method of ovary injection,while the highest transformation ratio were got by this approach.The injection of DNA solution at about 12 hours after pollination resulted in the highest transformation rate,while the concentration、pH value and the volume of DNA solution were important factors affecting fruit setting and seed production.GUS analysis and southern blotting hybridization results showed that NPTⅡ and GUS gene were transformed into cucumber.
