摘  要:目的探讨FHL1在低氧性肺动脉高压发病机制中的可能作用。方法将20只雄性Wistar大鼠分为2组,即10只低氧性肺动脉高压为实验组(以常压低氧建立大鼠肺动脉高压模型)和10只常压常氧正常对照组。右心导管法测定2组大鼠平均肺动脉压力(mPAP),并分别计算管壁厚度占血管外径的百分比(WT%)和管壁面积占血管总面积的百分比(WA%)。酶联免疫吸附法检测2组大鼠血清FHL1浓度和荧光PCR法观察大鼠肺组织FHL1 mRNA表达。结果实验组大鼠肺动脉压(2.86±0.39)kPa,右心室肥厚指数[RV/(LV+S)]为(43.53±3.38)%、WT%为(35.24±11.2)%,WA%为(55.09±12.38)%,分别与对照组的(1.35±0.28)kPa、(23.68±3.48)%、(23.63±9.74)%和(41.62±12.83)%比较明显升高(P<0.05);血清FHL1浓度(ng·mL-1)高于对照组[(646.2±41.8)ng·mL-1与(364.8±38.5)ng·mL-1,P<0.05],实验组大鼠肺组织FHL1 mRNA的含量亦较对照组增加[(8.47 e-3±3.12 e-3)与(5.407 e-3±2.31 e-3),P<0.05]。相关分析表明,FHL1 mRNA与WT%、WA%、mPAP呈正相关(P<0.05)。结论低氧后大鼠肺组织FHL1的合成和释放增多,FHL1增加可能与低氧性肺动脉高压的发生有关。Objective To evaluate the role of FHL1 (four and half LIM domainl, FHL1 ) in the pathogenesis of hypoxic pulmona- ry hypertension. Methods Twenty male Wistar rats were divided into two groups and exposed to air and isobaric hyp0xia for 3 weeks, re- spectively. The pulmonary artery pressure was measured by right cardiac catheterization. The rates of wall thickness/external diameter ( WT% ) and wall area/total vascular area ( WA% ) were calculated. The FHLI level in serum was measured by enzyme - linked immu- nosorbent assay. FHL1 mRNA expression in lung tissue were observed by PCR. Results The pulmonary artery pressure in the exposed rats was increased. The chronic hypoxia also elicited the thick of the wall and the narrowing of the lumen of pulmonary arterioles. It led to the in- creases of the pulmonary artery pressure, the index of right ventricular hypertrophy ( RV/( LV + S), the ratio of vascular wall thickness/vas- cular external diameter ( WA% ) and the ratio of vascular wall area/total vascular area ( WT% ) were compared with the controls PAP (2.86 ±0.39) kPa vs( 1.35 ±0.28) kPa;RV/( LV + S) :43.53 ± 3.38 vs. 23.68 ± 3.48 ;WT% : (35.24 ± 11.2) % vs(23.63±9.74)% ; WA% : (55.09 ± 12.38 ) % vs (41.62 ± 12.83 ) %, respectively,P 〈 0.05 ). In hypoxic group, the expression of FHL1 mRNA in the lung was significantly up - regulated,the expression level of FHL1 in serum was higher than that in control group ( P 〈 0.01 ). Linear correlation a- nalysis showed that FHL1 mRNA was positively associated with WF% ,WA% and mPAP (P 〈 0.05 ). Conclusion The FHL1 may play an important role in the pathogenesis process of hypoxic pulmonary vascular remodeling and pulmonary hypertension.
