摘  要:目的:探讨小泛素样修饰蛋白(SUMO)化修饰对核因子(NF)-κB通路激活的负性调控在2型糖尿病大鼠脑部病变中的干预作用,为前列地尔对其病变的早期干预提供依据。方法:随机抽取40周龄的10只GK大鼠为前列地尔组(前列地尔2.5μg/g,尾尖静脉注射给药,1次/d,共10d)、10只为糖尿病对照组,另取10只Wistar雄性大鼠作为正常对照组。正常对照组和糖尿病对照组给予等剂量生理盐水,均为尾尖静脉注射给药。三组均给予普通饲料。测定和观察血糖、甘油三酯(TG)、总胆固醇(TC)变化及NF-κB抑制因子(IκB)、NF-κB、SUMO1的水平变化。结果:(1)糖尿病对照组、前列地尔组大鼠的血糖、TG、TC均显著高于正常对照组(P均<0.05);(2)HE染色观察:正常对照组脑组织细胞形态正常,糖尿病对照组、前列地尔组脑组织可见细胞肥大,疏松样改变,前列地尔组病变较轻;(3)免疫组化测定的IκB、NF-κB、SUMO1:正常对照组少许阳性表达,糖尿病对照组、前列地尔组表达均显著增加(P<0.05或<0.01),但前列地尔组较糖尿病对照组IκB[(0.242±0.034)比(0.268±0.017)]、NF-κB[(0.373±0.015)比(0.486±0.013)]表达显著减少,SUMO1[(0.463±0.015)比(0.343±0.018)]表达显著增加(P均<0.05)。结论:小泛素样修饰蛋白在糖尿病大鼠脑组织表达增多,在前列地尔组表达更多,前列地尔对糖尿病大鼠脑组织病变有一定改善作用。Objective: To explore the intervention role of small ubiquitin related modifier protein (SUMO) on negative regulation activated by nuclear factor (NF) - κB in cerebral lesion of rate with type 2 diabetes mellitus (T2DM), and provide evidence for alprostadil early intervention on cerebral lesion in T2DM rats. Methods: A total of 10 GK rats of 40 - week old were randomly enrolled as alprostadil group (received alprostadil 2.5μg/g, via tail vein injection once/d for 10d), 10 GK rats were regard as DM control group, and another 10 Wistar male rats were regard as normal control group. Normal control group and DM control group received normal saline of the same dose via tail vein injection. All three groups were fed with normal diet. Changes of levels of blood,-gtucose, triglyceride (TG), total cholesterol (TC), expressions of IκB (inhibitor of NF- κB), NF- κB and SUMO1 were examined and observed in three groups. Results: (1) Compared with normal control group, there were significant increase in levels of blood glucose, TC and TG in DM control group and alprostadil group, P〈0.05 all; (2) HE staining observa- tion: cell form in cerebral tissues was normal in normal control group, while cell hypertrophy and porosity - like change were observed in cerebral tissues of DM control group and alprostadil group, but lesion of alprostadil group was significantly less than that of DM control group ; (3) Immunohistochemistry: IκB, NF- κB and SUMO were slightly positive expressed in normal control group, while expressions of them significantly increased in DM control group and alprostadil group (P〈0.05 or 〈0.01), but compared with DM control group, there were significant decrease in expressions of IκB [ (0. 268 ±0. 017) vs. (0. 242 ±0. 034)] and NF- κB [ (0. 486 ±0. 013) vs. (0. 373 ±0.015)], and significant increase in SUMO expression [ (0. 343 ±0. 018) vs. (0. 463 ±0. 015)] in alprostadil group, P〈0.05 all.
