摘 要:为研究J-亚群禽白血病病毒(ALV-J)在安徽省鸡群中的遗传变异趋势,对分离到的2株安徽省ALV-J毒株,克隆了其gp85基因序列,并与国内外分离毒株的相应基因序列进行了比较。结果显示,ALVJ gp85基因序列具有高度的易变性,可变氨基酸残基占全长序列的22.8%。其中高变区hr1和hr2具有较多的可变位点,而可变区vr2和vr3相对较为保守。二级结构预测显示ALV-J gp85蛋白含有大量的无规则卷曲结构,尤其是hr2区域。同源性分析表明,毒株AH1103和AH1104的gp85氨基酸序列同源性为95.8%;毒株AH1103与毒株BJ090201J,ev/J的gp85序列同源性最高,毒株AH1104的gp85序列则与国内毒株HuB09JY04的同源性最高。进化分析显示,毒株AH1103和AH1104均与国内毒株NX0101和SD07LK1及美国分离株4817的遗传距离较远。In order to further understand the genetic variation of subgroup J avian leukosis virus(ALV-J)in Anhui,ALV-J strains were isolated,and the gp85 gene was cloned by PCR.And we compared the amino acid sequences with thirteen sequences of ALV-J gp85 genes in the GenBank database.The results showed that ALV-J gp85 gene was highly variable,mutation sites accounted for 22.8% of full length amino acid sequence.hr1 and hr2 domains had high mutation frequency,while vr2 and vr3 domains were relatively more conservative.Protein secondary structure prediction showed ALV J gp85contained much random coil conformation,expecially hr2 domain.The homology between ALV-J gp85 amino acid sequences of strains AH1103 and AH1104 was 95.8%.The highest homology with strain AH1103 were strains BJ090201J and ev/J,and strain AH1104 had the highest homology with strain HuB09JY04isolated in China.Moreover,evolution analysis further revealed that the genetic distance strains AH1103 and AH1104 with strains NX0101,SD07LK1 and 4817 was farther.
