摘  要:为研究长春花生物碱(conophylline)诱导永生化猪胰腺源间充质干细胞(iPMSCs)对糖尿病小鼠的治疗效果,将长春花生物碱诱导的iPMSCs细胞移植到裸鼠体内2周后进行致瘤性安全检测;以双硫腙染色(DTZ)检测诱导iPMSCs的胰岛素功能性分化;将诱导的iPMSCs移植到糖尿病模型的小鼠体内,通过监测小鼠体重和血糖的变化,判断长春花生物碱诱导的iPMSCs对糖尿病小鼠的治疗效果。结果表明,诱导后的iPMSCs无致瘤性,呈DTZ阳性。诱导的iPMSCs移植到糖尿病小鼠体内后,与对照组小鼠相比,诱导的细胞会减缓糖尿病小鼠体重的降低趋势,且在短期内(1周)能恢复高血糖至正常水平。说明长春花生物碱具有诱导iPMSCs分泌胰岛素、短期治疗糖尿病的作用。The therapeutic effects in diabetic mice were observed to determine the inductive role of vinca alkaloids(conophylline)to immortal porcine mesenchymal stem cells(iPMSCs)from piglet pancreas.The iPMSCs were induced with combined culture media contained conophylline.The oncogenicity of induced iPMSCs was detected after induced iPMSCs were transplanted into nude mice 2weeks.The insulin functionality of induced iPMSCs was performed by Dithizone staining(DTZ).According to the changes of lost weight and plasma glucose concentrations of diabetic mice model,the therapeutic effects of induced iPMSCs on diabetic mice were estimated after induced iPMSCs were transplanted.The results suggested that inducted iPMSCs had no carcinogenic character,and DTZ positive.Compared with the control mice,the induced cells could slowed the lost weight in diabetic mice,and restored the high blood glucose concentration to normal in a short period of time(1week).The conclusion was that the induced iPMSCs by conophylline could secrete insulin and have the effect of treatment diabetes in short-term.
