梗死心肌骨骼肌卫星细胞移植后心肌细胞结蛋白变化

摘　　要：目的　观察骨骼肌卫星细胞梗死心肌移植后心肌细胞结蛋白的变化。　方法　自犬臀部取骨骼肌体外提取、培养、扩增骨骼肌卫星细胞 ,4′,6 -二脒基 - 2 -苯吲哚 (DAPI)荧光标记 ,经结扎的左冠状动脉前降支远端灌注移植入自体急性心肌梗死区域 ,2周、4周和 8周后将犬处死 ,取病理标本行免疫组织化学检测心肌细胞结蛋白表达、组织切片染色观察组织结构。　结果　在梗死区部位观察到带荧光的细胞核及肌纤维 ,部分已分化成横纹肌并以闰盘相连。正常区域心肌细胞结蛋白排列位于心肌细胞 Z线处 ,可见清晰显示的肌小节 ;梗死区心肌细胞结蛋白表达明显高于正常区域心肌细胞 ,但排列紊乱 ,整个心肌细胞均见阳性表达的结蛋白 ,未见肌小节 ;4周、8周时治疗组部分梗死心肌细胞已恢复排列并重见肌小节 ,但梗死区心肌细胞结蛋白表达仍明显高于正常区域心肌细胞 (P<0 .0 1)。　结论骨骼肌卫星细胞梗死心肌移植后对梗死心肌的组织结构起到了保护作用 。Objective\ To observe the changes of infarcted myocardial desmin after transplantation with skeletal muscle satellite cells.\ Methods\ Skeletal muscle satellite cells, that cultured and expanded in vitro, were labeled with 4′,6 diamidino 2 phenylindole and transplanted into the acute myocardial infarct site via the ligated left anterior descending artery. In the 2nd, the 4th, the 8th week, the dogs were sacrificed and the specimens were taken to examine.\ Results\ The skeletal muscle satellite cells with fluorescent label had been observed in the infarct site with consistent oriented direction similar to host myocardium. A portion of the transplanted cells had differentiated into muscle fiber with striation and connected with intercalated discs. Desmin was localized at the Z lines in normal myocardium, it was even possible to count the number of sarcomeres. In the infarct site, the expression of desmin was higher than that in normal myocardium, but the desmin was disorderly arranged and the sarcomeres disappeared. The same results could be seen in the 2nd week transplanted group similar as the control group. In the 4th, the 8th week, desmin in transplanted group was found to be normal and the sarcomeres appeared again partly, the expression of desmin in infarcted myocardium was also higher than that in normal site. Conclusion\ This study suggested that skeletal muscle satellite cells transplanted into the acute infarcted site, had the ability of structural preservation of the myocardial tissue, and induce normal distribution of myocardial desmin.
