HPLC-ELSD法测定清开灵滴丸中胆酸和猪去氧胆酸的含量

摘　　要：建立了高效液相色谱-蒸发光散射检测法（HPLC-ELSD ）测定清开灵滴丸中胆酸和猪去氧胆酸含量的方法．采用汉邦C18色谱柱（200mm×4.6mm ，5μm），流动相为0.1％甲酸-甲醇-乙腈溶液（25∶60∶15），柱温30℃，漂移管温度为105℃，空气流速2.2 L/min ．胆酸和猪去氧胆酸的进样浓度分别在0.598～1.596 mg/mL （ r＝0.9992）和0.600～0.1.600 mg/mL （ r＝0.9995）范围内其线性关系良好，胆酸和猪去氧胆酸平均回收率分别为99.74％和99.21％．该方法准确，简便，分离度高，重现性好，可用于清开灵滴丸中胆酸和猪去氧胆酸含量的测定．To establish the contents of Cholaiic Acud and Hyodeoxycholic Acid in Qingkailing Diwan by high per-formance liquid chromatography with evaporative light scattering detection （HPLC-ELSD ） .Methods：The analytical column was Hanbon Lichrospher C18 column （200mm × 4.6mm ,5μm）;mobile phase was 0.1% formic acid-meth-anol-acetonitrile （25∶60∶15 ）;column temperature of 30℃ ;drift tube temperature of 105℃ ,flow rate of the air at 2.2L/min .Results：A better linearity and good correlation coefficient were obtained Cholaiic Acud was 0.598~1.596 mg/mL （ r=0.9992 ） and Hyodeoxycholic Acid was 0.600~0.1.600 mg/mL （ r=0.9995 ） .Cholaiic Acud and Hyodeoxycholic Acid ,average recoveries were 99.74% （RSD=1.4% n=9 ） ,99.21% （RSD=1.0% n=9 ） . Conclusion ：The method is accurate ,simple ,Well-separated ,it can be used applicable Cholaiic Acud and Hyode-oxycholic Acid by Qingkailing Diwan .
