摘  要:目的探讨Ghrelin对内皮型一氧化氮合成酶(eNOS)活性的影响和作用机制。方法分离培养大鼠主动脉血管内皮细胞并分为对照组(c组)和Ghrelin组。Ghrelin组加入终浓度为10、50、100和200nmol/LGhrelin孵育3h收集标本。采用Westernblot法测定内皮细胞eNOS和PKC蛋白表达水平。结果与对照组相比,Ghrelin组eNOS磷酸化水平呈剂量依赖性增加(P〈0.05),PKC磷酸化水平呈剂量依赖性降低(P〈0.05)。结论Ghrelin可提高内皮细胞eNOS的活性,其机制可能与PKC通路的失活有关。Objective To investigate the effect of Ghrelin on endothelial nitric oxide synthase (eNOS) activity and its mechanism. Methods Vascular endothelial cells (ECs) were isolated from aorta in rats and were cultured. ECs were divided into two groups: control group and Ghrelin group. The cells were incubated with Ghrelin in different concen- tration ( 10, 50, 100 and 200 nmol/L) for 3 hour in Ghrelin group. The protein expression levels of nitric oxide synthase (eNOS) and protien kinase C (PKC) were measured by Western blot. Results Ghrelin can upregulate the phosphoryla- ted eNOS expression level, and downregulate signal conduction pathway PKC phosphorylation in a dose dependent manner ( P 〈 0. 05 ). Conclusions Ghrelin can improve the activity of eNOS in ECs, and it may be related to the loss of PKC ac- tivity.
