摘 要:目的分析高脂血症在兔心肌细胞增殖及凋亡中的作用。方法将普通级大耳白兔随机分为两组,每组10只。普通饲料组给予普通饲料,高脂饲料组给予高脂饲料,均喂养13w。实验前及实验第4、7、10和13周末分别取空腹兔耳缘静脉血测血脂水平;实验结束光镜下观察心肌细胞的病理形态学改变,并采用FCM观察各组心肌细胞增殖与凋亡的情况。结果第4、7、10及13周末,高脂饲料组血清TC、TG、HDL-C和LDL-C水平均显著高于普通饲料组(P<0.01)。普通饲料组心肌细胞结构基本正常,高脂饲料组心肌细胞出现空泡变性、心肌弥漫性纤维化和淋巴细胞浸润。高脂饲料组G0/G1期百分率较普通饲料组显著增高(P<0.05),而S期、G2/M期百分率及PI均较普通饲料组显著降低(P<0.05或P<0.01)。结论高脂血症可引起兔心肌细胞病理性改变,抑制心肌细胞增殖,且对其凋亡无显著影响。Objective To investigate the effect of hyperlipidemia on the proliferation and apoptosis of eardiomyocytes of rabbits. Methods The ordinary-grade white rabbits were randomly divided into 2 groups, 10 rabbits in each group. The normal diet group and high-fat diet group were respectively fed with normal feed and high- fat diet for 13 weeks. The fasting venous blood from the ear edge was drawn to measure the level of blood lipids before experiment and at the end of weeks 4,7,10 and 13 of experiment. After experiment,the pathological changes of myocardial tissue were detected by light microscope,and the proliferation and apoptosis of cardiomyocytes were observed by flow cytometry(FCM). Results At the end of weeks 4, 7, 10 and 13, the levels of serum total cholesterol ( TC ), triglyceride ( TG), high-density lipoprotein cholesterol ( HDL-C ) and low-density lipoprotein chotesterol(LDL-C)in the high-fat diet group were atl significantly higher than in normal diet group(P〈0.01). The structure of cardiomyocytes was normal in normal diet group, but in high-fat diet group, the cardiomyocytes appeared vacuolar degeneration, myocardial diffuse fibrosis and lymphocytic infiltration. The percentage at G0/G1 phase in high-fat diet group was obviously higher than in normal diet group(P〈0.05), whereas those at S phase,G2/M phase and proliferation index(H)were markedly lower than in normal diet group(P〈0.05 or P〈0.01). Conclusion Hyperlipidemia can induce the pathological changes of cardiomyocytes in rabbits, inhibit its cardiomyocytes proliferation and has no obvious effect on the apoptosis of cardiomyocytes.
