摘  要:目的研究低频超声对大分子中药的经皮促渗作用,为超声透药方法在临床中的应用提供实验依据。方法选用家兔为研究对象,通过离体实验和在体实验2个实验观察低频超声对5%桂枝萃取液的促透作用。离体实验分为对照组与超声组,对照组采用自然渗透法,超声组采用超声促透中药法,采用高效液相色谱法(HPLC)对不同观察时间点的桂皮醛含量进行检测,并将2组的检测结果进行统计学分析;在体实验同样分为对照组与超声组,对照组采用中药热敷法,超声组采用超声促透中药法,采用HPLC法对不同组织(关节液、滑膜、肌肉、皮肤)中的桂皮醛含量进行测定,并将两组检测结果进行统计学分析。结果离体实验中药物浓度随时间的延续而增加,不同时间点超声组浓度较高,超声组和对照组差别有统计学意义(P<0.05);在体实验中关节液、滑膜、肌肉、皮肤中均检测到桂皮醛的含量,超声组明显高于对照组(P<0.05)。结论低频超声可提高皮肤的通透性,促进中药大分子物质的通过,值得临床进一步研究。Objective To study the effect of low-frequency ultrasound for enhancing percutaneous permeation of Chinese medicinal with macromolecule, and provide the experimental evidence for the clinical application of phonophoresis. Methods Taking rabbits as objects, the effect of enhancing permeation of low-frequency ultrasound for 5% extract liquor of Guizhi (Ramulus Cinnamomi ) was observed through experiment in vitro and experiment in vivo. In experiment in vitro, rabbits were divided into control group in vitro and ultrasound group in vitro. The control group in vitro was give nature penetration method and ultrasound group in vitro, phonophoresis. The content of cinnamic aldehyde was detected by using high performance liquid chromatography (HPLC) at different time points and the outcomes from two groups were statistically analyzed. In experiment in vivo, rabbits were divided into control group in vivo and ultrasound group in vivo. The control group in vivo was give hot compress of Chinese medicinal and ultrasound group in vivo, phonophoresis. The content of cinnamic aldehyde in different tissues (including synovial fluid, synovium, muscle and skin) was detected by using HPLC and the outcomes from two groups were statistically analyzed. Results The content of cinnamic aldehyde in experiment in vitro increased over time, which was higher in ultrasound groups at different time points. The difference between ultrasound groups and control groups had statistical significance ( P 〈 0. 05 ). In experiment in vivo, cinnamic aldehyde was detected in synovial fluid, synovium, muscle and skin, and its content was higher in ultrasound group in vivo than that in control group in vivo (P 〈 0.05). Conclusion Low-frequency ultrasound can improve the permeability of skin and enhance the permeation of Chinese medicinal with macromolecule. It is worth further studing in clinic.
