慈菇消脂丸对酒精性脂肪性肝病大鼠血清APN与TNF-α的影响

摘　　要：目的研究慈菇消脂丸对酒精性脂肪性肝病(AFLD)大鼠血清脂联素(APN)与肿瘤坏死因子(TNF-α)的影响,探讨其作用机制。方法用高脂饲料喂食加乙醇灌胃的方法复制酒精性脂肪肝模型。随机分为正常组、模型组、慈菇消脂丸高剂量组、中剂量组、低剂量组、罗格列酮阳性药物对照组。慈菇消脂丸对模型大鼠治疗4周后,观察模型大鼠肝组织病理形态学的变化;血清肝功能、APN和TNF-α含量变化。结果模型组大鼠肝小叶内出现大量脂肪变性,汇管区伴有炎性细胞浸润以及部分点状肝细胞坏死。与正常组相比较,模型组大鼠谷丙转氨酶(ALT)、谷草转氨酶(AST)、TNF-α均升高,APN明显下降。与模型组相比较,慈菇消脂丸各剂量组,肝脏脂肪变性和炎症均有不同程度的减轻。慈菇消脂丸能降低模型大鼠ALT、AST、TNF-α含量,对APN亦有较为明显的改善,差异具有统计学意义(P<0.05)。结论慈菇消脂丸具有改善肝功能,调节TNF-α和APN这2种脂质因子,减轻肝细胞脂肪变性,从而起到对酒精性脂肪性肝病的治疗作用。Objective： To study the effect of Cigu Xiaozhi Wan on serum adiponectin （APN） and tumor necrosis factor （TNF-α） in rats with alcoholic fatty liver disease （AFLD）, and to discuss the mechanism. Methods The rat model of AFLD was established by feeding high fat diet and intragastrically giving ethanol. All rats were randomly divided into normal group, model group, high-dose group, mid-dose group, low-dose group and positive control group. After 4 weeks of treatment with Cign Xiaozhi Wan on model rats, histopathological changes of liver were observed; serum liver function and levels of APN and TNF-α were detected. Results In model group, there were steatosis in hepatic lobule, infiltration of a large number of inflammatory cells and spotty necrosis of partial hepatocytes in portal area. The levels of ALT, AST and TNF-ot increased and the level of APN decreased significantly in model group compared with those in normal group. In high-dose, mid-dose and low-dose groups, steatosis and inflammation in liver were relieved in varying degrees. Cigu Xiaozhi Wan could decrease the levels of ALT, AST and TNF-α of model rats, and ameliorate the level of APN significantly, the differences of which were statistically significant （ P 〈 0.05 - 0.01 ）. Condusion Cigu Xiaozhi Wan has the effects of ameliorating liver function, regulating the levels of TNF-α and APN and relieving steatosis of hepatocytes, so as to be used in treatment on AFLD.
