摘 要:本实验能研究猪CD4基因启动子多态性、基因表达在不同品种间的差异。实验采用PCR-RFLP和实时定量PCR技术,分析大白、长白猪CD4基因启动子多态性及外周血CD4基因相对表达量。结果表明:实验的2个群体中,CD4基因启动子区存在一个SNP位点(-152A→T),经Apo I酶切后得到AA、AT、TT 3种基因型,T均为优势等位基因;经χ2适合性检验,大白、长白2个群体都不处于Hardy-Weinberg平衡状态,这2个品种的基因型频率分布存在显著性差异(χ2=7.59,P<0.05),基因频率分布差异不显著(χ2=0.87,P>0.05);大白猪的外周血CD4基因表达量极显著高于长白(P<0.01)。本研究为后续猪CD4基因多态性与抗病性状的关联分析奠定基础。There are many reports about polymorphism analysis of CD4 gene in humans and other animals, but no in swine. The polymorphism in the promoter of porcine CD4 gene (-152A→T) was detected by PCR-RFLP within ApoI, and the difference of CD4 gene expression in breeds was analyzed by real-time PCR in Yarkshire and Landrace. The results showed that there were three genotypes, AA, AT, Tr. T was the predominant allele; By the x2test, those genotypes didn't fit the Hardy-Weinberg balance. There was significant difference among genotype frequencies in the two species (X2= 7.59, P〈0.05), no difference in allele frequency (X2=0.87, P〉0.05). CD4 gene expression in peripheral blood of Landrace is significantly lower than that of Yarkshire (P〈0.01). It could provide the basis for the association analysis between the SNPs of CD4 gene and disease resistance trait.
