摘 要:目的通过测定染氟大鼠血清中骨形成抑制蛋白(DKK1)及核心结合因子(Runx2)、碱性磷酸酶(ALP)的表达,探讨氟通过调控DKK1对大鼠成骨作用的影响。方法 SPF级成年SD大鼠64只,随机分为对照组和3个染氟组,氟化钠(NaF)溶液饮水染毒,浓度分别为1.6、16、32mg/kg NaF。分别于染氟第0、第35、第90天时,大鼠尾动脉取血,ELISA法测定各时段血清中DKK1及第90天时Runx2浓度,全自动生化分析仪测定ALP活力水平。染毒末期处死动物,取双侧股骨骺端组织行HE染色。结果 HE染色显示高剂量组骨皮质及骨膜均增厚,中剂量组骨小梁增生、增宽,间隙减小;染氟第90天时,中、高剂量组DKK1浓度低于对照组(10.43±1.04μg/L),差异有统计学意义(F=23.05,P<0.05),且各染氟组DKK1浓度较第0天时明显降低,差异有统计学意义,(t低=2.65,t中=13.36,t高=8.40,P<0.05);染氟第90天时,染氟组Runx2浓度均高于对照组(122.12±2.46μg/L),差异有统计学意义(F=10.61,P<0.05),ALP活力均高于对照组(183.67±4.30U/L),差异有统计学意义(F=35.04,P<0.05);DKK1浓度与Runx2浓度及ALP活力呈显性负相关(r=-0.34,r=-0.47,P<0.05)。结论氟致大鼠血清中DKK1表达降低,并以此促进大鼠的成骨作用。Objective To explore the effects of fluoride regulated DKK1 expression on osteogenesis of rats through analyzing the changes of DKK1 and Runx2, ALP expression in serum of fluoride exposed rat. Methods 64 adult SD rats were randomly divided into 3 fluoride exposed groups(1.6,16,32 mg NaF/kg)and a control group. Caudal artery blood samples of rats were taken on the 0, 35th and 90th day, serum DKK1 of various periods and Runx2 concentrations at the 90th day were analyzed by ELISA. Levels of ALP activities were determined by automatic biochemistry analyzer. On the end of exposure double sides of femoral epiphysis were sectioned and stained by HE. Results HE stained sections showed the thickening of periosteum in high --dose group, and broadening of bone trabecula in mid--dose group. The concentrations of DKK1 of mid-- and high--dose groups on the 90th day exposure were significantly lower than those of the control(F= 23.05, P〈0. 05). The levels of DKK1 of all 3 fluoride exposed groups were significantly lower than that of the 0 exposure(tlow= 2. 65, tmid= 13.36, thigh: 8. 40, P〈0.05). On the day of 90th exposure, the levels of Runx2 and ALP activities of all 3 exposed groups were significantly higher than those of the control(F: 10. 61,F = 35.04, P 〈:0.05). The concentrations of DKK1 and Run2 were significantly and negatively correlated with the activity of ALP (r = - 0. 34, r =-- 0.47, P 〈0.05). Conclusions Fluoride induces the decrease of the expression of serum DKK1, which promotes the osteogenesis of rats.
