摘 要:目的探讨白藜芦醇对过氧化氢(H2O2)诱导鼠晶状体上皮细胞凋亡损伤的保护作用。方法取健康SD大鼠透明晶状体60只,随机分为5组,每组12只,分别给予高、中、低剂量(5、10、20μmol/L)的白藜芦醇和100μmol/L H2O2,利用透射电镜观察超微结构变化并测定凋亡因子caspase-3及caspase-9的表达。结果伴随白藜芦醇给药浓度的增加,透射电镜下鼠晶状体上皮细胞损伤的程度减轻,此外,白藜芦醇对caspase-3及caspase-9的表达具有抑制作用。结论白藜芦醇可以抑制H2O2诱导的晶状体上皮细胞的凋亡,对氧化损伤性白内障具有预防作用,为寻求有效防治白内障药物提供了可靠的实验依据。Objective To discuss the protective effect of resveratrol on human lens epithelial cells (HLEC) against H2O2-induced apoptosis.Methnds Sixty lenses of healthy SpragueDawley (SD) rats were randomly devided into 5 groups,then incubated with different concentration of resveratrol (5,10,20 μmol/L) and H2O2,ultrastructure changes were observed,and apoptosis expression of caspase-3 and caspase-9 were assayed under transmission electron microscope.Results The damage of human lens epithelial cells were alleviated with resveratrol concentration elevated,and the expression of caspase-3 and caspase-9 was also inhibited.Conclusion Resveratrol may inhibit H2O2-induced apoptosis of HLEC,and has protective function against oxidative damage cataract.It provides reliable evidence for pursuing effective medicine to prevent and treat cataract.
