熊胆眼药水滴眼后熊去氧胆酸在眼内基本分布的实验研究

摘　　要：目的探讨熊胆眼药水滴眼后其标志性成分熊去氧胆酸(UDCA)在家兔眼球的房水与玻璃体中分布及其穿过眼表屏障进入房水及玻璃体的能力。方法将48只家兔随机分为6组,每组8只。将家兔双眼各点5次熊胆眼药水,点眼后第1～5组于0.5 h、1 h、2 h、4 h、8 h时左眼取玻璃体,右眼取房水,第6组为对照组。用高效液相色谱仪测量每个样本中UDCA的含量。结果第1～5组,家兔双眼点5次熊胆眼药水后,0.5 h、1 h、2 h、4 h、8 h,房水样品处理后的UDCA的浓度均值分别为:1.21μg/mL、3.53μg/mL、33.68μg/mL、37.36μg/mL、39.12μg/mL。玻璃体样品处理后UDCA浓度分别为4.20μg/mL、13.20μg/mL、17.72μg/mL、21.48μg/mL、23.64μg/mL。结论熊胆眼药水滴眼后能通过眼表屏障进入房水及玻璃体,进入时间越长到达的能力越大。Objective To investigate the distribution of marking composition ursodeoxycholic acid in aqueous and vitreous,in addition ability of penetraing ocular surface barrier into the aqueous and vitreous humor after pointing bear bile eye drops to eyes.Methods Forty-eight rabbits were randomly divided into six groups,and each group of eight rabbits.Bear bile eye drops pointed both eyes of rabbits five times.After that,we collected vitreous of rabbits＇ left eyes and aqueous of rabbits＇ right eyes from the first group to the fifth group at 0.5 h,1 h,2 h,4 h,8 h.The sixth group was control group.Ursodeoxycholic acid content of each samples was measured by using high performance liquid chromatograph.Results After pointing bear bile eye drops to both eyes of rabbits for five times,the mean concentration of UDCA in the aqueous sample were：1.21 μg/mL,3.53 μg/mL,33.68 μg/mL,37.36 μg/mL,39.12 μg/mL,respectively from the first group to the fifth group at 0.5 h,1 h,2 h,4 h,8 h.The concentration of UDCA in vitreous samples were 4.20 μg/mL,13.20 μg/mL,17.72 μg/mL,21.48 pg/mL,23.64 μg/mL,respectively.Conclusion Bear bile eye drops can penetrate the ocular surface barrier into the aqueous humor and vitreous after pointing eyes.The longer to arrive,the more ability when arrive.
