中药罗仙子抗炎活性部位对TNF-α诱导的人脐静脉内皮细胞增殖及Matrigel小管形成的影响

摘　　要：目的以肿瘤坏死因子-α(TNF-α)诱导的人脐静脉内皮细胞(HUVEC)为研究对象,探讨罗仙子(Musca Domestica)抗炎活性部位在体外对HUVEC增殖及Matrigel小管形成的影响。方法将构建的HUVEC细胞分为正常对照组、模型组及罗仙子活性部位处理组,制备罗仙子抗炎活性部位,采用流式细胞术结合ModFit LT软件分析,明确罗仙子抗炎活性部位对HUVEC增殖的影响;采用Matrigel小管形成实验结合Image Pro Plus软件分析,明确罗仙子抗炎活性部位对HUVEC血管形成的影响。结果相对模型组,罗仙子抗炎活性部位处理组HUVEC增殖得到显著的抑制,增殖系数降低明显(P<0.01),同时,该处理组Matrigel小管形成的数量、面积和长度显著减少(P<0.01)。结论罗仙子抗炎活性部位对TNF-α诱导HUVEC增殖及Matrigel小管形成具有抑制作用。Abstract： Objective To investigate theon proliferation and Matrigel tube formation in human umbilical veinendothelial cells （HUVECs）,the TNF--a --induced HUVECs were introduced. Methods HUVECs induced by TNF were divided into normal con- trol group, model group and luoxianzi anti--inflammatory parts treatment group. Luoxianzi anti--inflammatory parts were harvested in accordance with the previous study. Flow cytometry combined with ModFit LT soft- ware analysis were used to define the effect on the HUVEC proliferation. And Matrigel tubule formation assay combined with Image Pro Plus software analysis were used to clarify the effect on the HUVEC angiogenesis.Results Compared to model group,HUVEC coefficient decreased significantly （ P 〈0.01） cell proliferation was significantly inhibited, and the proliferation in luoxianzi anti--inflammatory parts treatment group, while the Matrigel tubule formation number,size and length reduced significantly （ P 〈0.01）. Conclusion The luox ianzi anti--inflammatory parts has the function of inhibiting the proliferation and tubule formation of TNF--a -- induced HUVECs.
