摘  要:摘要:目的观察壮精合剂对初老大鼠学习记忆能力和海马区神经营养因子的影响,探讨其抗衰老的作用机制。方法选用10~12月龄,体重(300±20)g初老大鼠50只,随机分为5组,空白对照组,壮精合剂低、中、高剂量组,阳性药对照组,每组10只。各组分别灌胃相应药物8周后,采用Morris水迷宫检测各组大鼠的学习记忆能力和采用酶联免疫吸附实验双抗体夹心法,检测初老大鼠大脑海马内神经生长因子(NGF)、脑源性神经营养因子(BDNF)含量的变化。结果与空白对照组比较,壮精合剂高、中剂量组及阳性药对照组均能提高初老大鼠学习记忆能力及提高其海马内NGF和BDNF含量(P〈O.05),且壮精合剂高剂量组作用最为显著(P〈0.01)。壮精合剂高剂量组在提高初老大鼠学习记忆能力及提高其海马内NGF和BDNF含量方面与阳性药对照组比较差异有统计学意义(P〈0.05),且在提高BDNF含量方面差异有统计学意义(P〈O.01)。结论壮精合剂能有效提高初老大鼠的学习记忆能力,其作用机制可能与调节海马内NGF和BDNF含量有关。Abstract: Objective To observe the effects of Zhuangjing mixture (ZJM) on learning memory abilities and nerve nutritional factor in primary senile rats' hippocampus, and to study its meehdnism of anti--aging. Methods Fifty primary senile female rats aged months and weighed g were randomly divid- ed into 5 groups: blank control group, low--, mid-- and high--dose group of ZJM, positive control group, with 10 rats in each group. After medication for 8 weeks, Morris water maze test had been performed to assessthe learning and memory abilities of primary double antibody sandwich method was used to rived neurotrophic factor (BDNF) in primary senile rats and enzyme--linked immunosorbent assay (ELISA) detect the content of nerve growth factor (NGF) and brain--de senile rats hippocampus. Results Compared with blank con trol group, high--, mid-- dose ZJM group and positive drug control group could improve the learning and memory abilities of primary senile rats and increase the content of NGF and BDNF in the hippocampus ( P 〈 0.05), and high--dose ZJM group had the best results ( P〈0.01). Compared with the positive drug control group, high--dose ZJM group had higher learning and memory abilities and higher content of NGF and BDNF in the hippocampus, there were significant differences ( P 〈0.05), and they had highly significant differences in the content of BDNF ( P 〈0.01). and memory ability, and its mechanism Conclusion may be re ZJM can effectively improve the primary senile rat learning lated to regulation of NGF and BDNF in hippocampus.
