半枝莲多糖对S180荷瘤小鼠脾淋巴细胞内Ca^2＋浓度的影响

摘　　要：本文通过观察荷瘤小鼠淋巴细胞内钙离子浓度的变化,初步探究Ca2+作为第二信使在半枝莲多糖诱导淋巴细胞增殖过程中调控作用。采用荧光探针Fura-3/AM对细胞内的Ca2+进行标记,在激光共聚焦显微镜下观察细胞的荧光强度变化。结果表明,半枝莲多糖各剂量组与空白对照组比较均对脾淋巴细胞中钙离子的浓度有显著提高作用(P<0.01),并呈现剂量依赖关系,而与阳性组比较各剂量组均低于阳性组,低、中剂量组有差异(P<0.05)。从试验结果看出,胞内Ca2+浓度的提高能促进淋巴细胞的增殖。Regulation and control function of Ca^2 ＋ as a second messeger in the process of Scutellaria barbata D. Don polyaccharides inducing lymphocyte proliferation were initially studied by observing the changes of calcium ion consistency within tumor-burdened mice lymphocytes. Fluorescent probe Fura-3/AM was used to mark Ca^2＋ in lymphocytes, and the changes of its fluorescence intensity were observed with the laser confocal microscope. The result showed that compared with the blank dose group, each Scutellaria barbata D. Don polyacchafides dose group had the significant improvement effect for calcium ion consistency in the splenic lymphocytes（ P 〈0.01 ） and performed a dose-dependent manner, however, it was low, compared with the positive group. The results of the treatments with low and middle dose groups had significant differences with that of control （ P 〈 0.05 ）. It was concluded that the improvement of Ca^2＋＋ consistency in lymphecytes would induce the proliferation of lymphocytes.
