摘 要:目的探索Dlx1的天然反义转录物(Dlx1as)在小鼠大脑发育过程中的功能。方法根据生物信息学分析与分子生物学实验验证Dlx1as是否具有多聚腺苷酸尾,采用实时定量PCR检测Dlx1as与Dlx1 mRNA在小鼠大脑发育过程中随时间的变化,利用原位杂交的技术比较Dlx1as与Dlx1 mRNA空间表达谱。结果 Dlx1as具有多聚腺苷酸尾,在小鼠的胚胎发育过程中,从E14.5到E18.5高表达,与Dlx1 mRNA在各时间点表达水平虽比较接近,但在表达区域存在互补的特点。结论 Dlx1as在小鼠大脑发育过程中有一段时间的高表达,与蛋白编码基因Dlx1 mRNA空间互补,推测其可能是通过调控Dlx1 mRNA来影响小鼠的大脑发育。Objective To explore the role of Dlxl as, a natural antisense transcript, in mice brain de- velopment. Methods Based on the bioinformatic and molecular biological findings, we confirmed whether the Dlxl as has a poly-A tail. The temporal expressions of Dlxl as and Dlxl mRNA were determined by quantitative re- al-time PCR and the spatial expressions of Dlxlas and Dlxl mRNA were compared by using in situ hybridiza- tion. Results Dlxl as had a poly-A signal. Dlxl as' s expression was at a high level through a period of time in the embryo development, which was similar with Dlxl mRNA in temporal but different in spatial. Conclusions Dlxlas is highly expressed in a certain period during mice brain development, which spatially supplements the mRNA expression of D1 xl. Therefore, it is supposed that Dlxl as influences the mice brain development by regu- lating Dlxl mRNA.
