摘 要:目的探讨鞘氨醇激酶-1(sphingosine kinase 1,SphK1)促进胃癌细胞转移和侵袭的机制。方法使用转染、RNA干扰及Sphk1过表达载体构建的方法分别实现胃癌细胞SGC-7901中SphK1蛋白的过表达、低表达,通过Transwell实验检测SphK1对胃癌细胞转移和侵袭能力的影响,并利用ELISA检测1-磷酸鞘氨醇(sphingosine 1-phosphate,S1P)及其受体水平的变化。结果 SphK1的过表达可显著促进胃癌细胞的转移和侵袭(P<0.05),而SphK1特异性siRNA可轻微降低胃癌细胞的转移和侵袭能力,但与对照相比差异不显著。SphK1转染细胞S1P表达水平(0.32±0.01)高于对照组(0.24±0.02)(P<0.05),SphK1的过表达可提高S1P的水平,而外源性S1P的添加可通过EDG1促进胃癌细胞的转移和侵袭。结论 SphK1/S1P/EDG1途径在胃癌的转移中有重要的作用,为胃癌的诊断及治疗提供了一个可能的新靶点。Objective To investigate the underlying mechanism of sphingosine kinase 1( SphK1) in promoting metastasis and invasion in gastric cancer cells. Methods Transfection,RNA interference and recombinant plasmid encoding SphK1( pWPXL-SphK1) were applied to change the expression of the target protein in gastric cancer cell line SGC-7901. Transwell assay was adopted to detect the metastasis and invasion ability of different types of SGC-7901 cells. The content of sphingosine 1-phosphate( S1P) in the supernatant was measured by ELISA. The mRNA expression of S1P receptors,EDG1 and EDG2,were studied by qRT-PCR. While the protein expression of SphK1,EDG1 and EDG2,was measured by Western blotting. Results SphK1 over-expression in SGC7901 cells enhanced the metastasis and invasion in the cells,while,SphK1 specific siRNA transfection reduced both the metastasis and invasion,without significant difference when compared with original SGC-7901 cells. Furthermore,overexpression of SphK1 also improve the expression of S1P,and exogenous S1P improved the migration and invasion in gastric cancer cells through EDG1 receptors. Conclusion SphK1 / S1P / EDG1 plays important roles in the metastasis in gastric cancer,and might provide new target for the diagnosis and therapy of the disease.
