摘  要:目的观察广西眼镜蛇毒蛋白Natrin在体外对人肝癌SMMC-7721细胞的增殖抑制作用及凋亡诱导作用。方法以不同浓度的Natrin作用于SMMC-7721细胞24 h后,用MTT法检测细胞的生长抑制率;AO/EB双染色法、透射电子显微镜法观察细胞凋亡、流式细胞仪检测细胞凋亡率及细胞周期。结果 Natrin对人肝癌SMMC-7721细胞有体外抑制作用,随着药物浓度的增大抑制率也随之增大,24 h半数抑制浓度IC50为138.69 mg·L-1;给药后,荧光显微镜下与透射电子显微镜下细胞呈现典型的凋亡形态改变;流式细胞仪检测到随着药物浓度的增大凋亡百分数也随之增大;Natrin能诱导SMMC-7721细胞发生S期和G2/M期阻滞。结论Natrin在体外能抑制人肝癌SMMC-7721细胞增殖并诱导其凋亡。Aim To observe the growth inhibitory effects and apoptosis inducing effects of Natrin on hu- man hepatocarcinoma SMMC-7721 cells in vitro. Methods The growth inhibition ratio of SMMC-7721 cells after exposure to different concentrations of Natrin was measured using MTY assay; morphological changes of apoptosis were detected by AO/EB staining and transmission electron microscopy; the cell cycle and apoptotic percentage were detected using FCM. Re- sults The growth of human hepatocarcinoma SMMC- 7721 cells was inhibited by the treatment of Natrin. With the increase of drug concentration, the growth in- hibitory rate also increased. ICso of 24h was 138. 69 mg · L^-1. After treated by Natrin, typical morphologi- cal changes of apoptosis were observed by fluorescence microscope and transmission electron microscope. With the increase of drug concentration, apoptotic percent- age performed by flow cytometry also increased. Natrin showed the effects of S and G2/M cell cycle arrest. Conclusion Natrin can inhibit proliferation and in- duce apoptosis in human hepatocarcinoma SMMC-7721 cells in vitro.
