摘  要:目的研究模拟失重效应条件下大鼠肌组织机械生长因子(mechano-growth factor,MGF)的表达变化,探讨MGF在失重性肌萎缩中的作用。方法采用大鼠尾部悬吊模型模拟失重效应,尾吊1、4、7、10、14天后,分离并分析肌肉相对重量变化,并通过实时定量PCR分析模拟失重效应对比目鱼肌(soleus,SOL)与趾长伸肌(extensor digitorum longus,EDL)中胰岛素样生长因子(insulin-like growth factor,IGF-I)两个选择性剪接体MGF、IGF-IEa及M-钙粘附分子(M-cadherin,M-Cad)mRNA表达水平的影响,同时分析IGF-IEa和MGF在两种肌肉中的表达差异。结果尾吊4 d后,SOL相对重量开始显著下降,而EDL相对重量只有轻微下降趋势。尾吊不同时间后,比目鱼肌的MGF、IGF-IEa和M-Cad等基因表达都表现出显著性下降;而在趾长伸肌中,只表现出轻微的下降趋势。结论在尾部悬吊处理条件下,不同肌肉类型MGF的表达变化不同。推测MGF表达的减少与SOL失重性肌萎缩的发生有关。: Objective To explore the effects of simulated microgravity on the expression of mechano-growth fac- tor(MGF) and try to understand the important role of MGF in muscle atrophy caused by microgravity. Meth- ods Hindlimb suspension (HS) model was used to simulate microgravity in rat. After 1, 4, 7, 10 or 14 days of HS, the muscles of soleus(SOL) and extensor digitorum longus(EDL) were separated and weighed. Then the effects of simulated weightlessness on the mRNA expression of MGF, IGF-IEa and M-Cad, as well as the relative change in expression of IGF-IEa and MGF in both muscles were analyzed with real-time quantitative PCR. Results After 4 days of HS, the relative mass of SOL began to decline significantly, while the relative weight of EDL was only shown a slight downward trend. At different time of HS, expression of MGF, IGF-IEa and M-Cad in SOL decreased significantly, but it didn't occurr in EDL. Conclusions The expression of MGF changed differently in different muscle types under the conditions of stimulated weightlessness. It is suggested that the reduction of MGF expression is related to the occurrence of muscle atrophy in SOL under microgravity.
