摘　　要：目的:采用大鼠灌胃给药的方法考察枳壳中柚皮苷和新橙皮苷在大鼠体内的分布特征。方法:采用色谱柱Diamonsil C18(150 mm×4.6 mm,5μm),流动相为乙腈-0.1%甲酸水(20∶80),流速1.0 mL·min-1,柱温25℃,检测波长283 nm,进样量20μL,大鼠组织匀浆液经乙腈沉淀蛋白,氮吹处理甲醇定容后进行HPLC分析。结果:大鼠单次口服给药60 min后,柚皮苷和新橙皮苷浓度在各组织中均达到最大值,其中小肠和胃中的浓度非常高,其次是肺、脂肪、肌肉,肾、脾和脑,二者在肝和心的浓度较低;240 min后,柚皮苷和新橙皮苷在心、脾、肾、肌肉等组织浓度很低甚至没有。结论:柚皮苷和新橙皮苷在大鼠体内的分布相对广泛,胃和小肠是其主要分布器官,容易在胃和小肠中蓄积,其他组织中分布较少。Objective： To determine the distribution Methods： The chromatographic conditions were as follows： of naringin and neohesperidin in rat tissues by HPLC separation was performed on Diamonsil Cls column （ 150 mm 4.6 mm,5 txm） with column temperature set at 25 ~C. The mobile phase was composed of acetonitrile- 0.1% formic acid （20： 80）. The flow rate was 1.0 mL min-1, and the detection wavelength was set at 283 nm. The injection volume was 20 ixL. The supernatant of tissue homogenate was evaporated to dryness under nitrogen at- mosphere after precipitating the tissue protein with aeetonitrile, and the residue was dissolved in methanol. Re- suits： At 60 min after single dose oral administration, the contents of naringin and neohesperidin in various tissues reached the maximum. The contents of naringin and neohesperidin in small intestine and stomach were the highest, followed by lung, fat, muscle, kidney, spleen, brain, liver and heart. At 240 min, the contents of naringin and neohesperidin in tissues including heart, spleen, kidney and muscle decreased significantly or could not be detec- ted. Conclusion： Naringin and neohesperidin could be detected in all the tissues, and stomach and small intestine were the major distribution tissue. Naringin and neohesperidin accumulated more in stomach and small intestine and less in other tissues
