摘 要:以猪去氧胆酸为原料,合成了鹅去氧胆酸与熊去氧胆酸。为优化中间体与目标产物的合成工艺,探索了反应条件及重结晶条件等。鹅去氧胆酸和熊去氧胆酸的总产率分别达到26%、19%,HPLC检测其纯度相应为97.5%、96.2%。通过熔点、MS和1 HNMR测定确证了各关键中间体及目标产物结构。改进后的工艺原料易得、总产率较高、后处理操作简便,具有较高的应用价值。In this paper, chenodeoxycholic acid(CDCA) and ursodeoxycholic acid(UDCA) were synthesized with hyodesoxycholic acid(HDCA) as starting material. To optimize the synthetic process of intermediates and target compounds, the reaction conditions and recrystallization conditions were investigated. The overall yield of CDCA and UDCA reached 26% and 19~ and their purity was 97.5% and 96.2% determined by HPLC,respec- tively. The structure of key intermediates and target compounds were confirmed by melting point, MS and 1 HNMR. The improved synthetic process had the advantages of easy availability of starting materials,high ste- reoselectivity,easy after-treatment and good overall yield,so it had considerable practical value.
