摘 要:以水栀子为原料,研究栀子多糖的闪式提取工艺。在单因素试验基础上,考察提取时间、乙醇体积分数和液料比对栀子多糖得率的影响,根据Box-Benhnken中心组合试验设计原理,采用响应面法对闪式提取条件进行优化。结果表明:栀子多糖的最佳提取工艺条件为:提取时间42 s液料比27∶1(mL∶g)、乙醇体积分数5%,在此条件下多糖得率为6.38%,与模型预测基本相符。与传统热水浸提法相比,闪式提取栀子多糖得率提高了32.5%,是较为理想的栀子多糖提取方法。Homogenate extraction conditions of polysaccharides from Gardenia jasminoides Ellis were studied.Single factors including extraction time,alcohol concentration,and liquid to solid ratio were investigated separately.The response surface analysis method followed by verification test was conducted for obtaining the appropriate operation conditions.Results showed that the optimal conditions of polysaccharides extraction were as follows:extraction time 42 s,alcohol concentration 5% and liquid to solid ratio 27∶1 (mL∶ g).Under the above conditions,the yield of polysaccharides was 6.38%.It matched with the predicted yield.Yield of polysaccharides using homogenate extraction was 32.5% higher than that in traditional heat refluxing extraction.Hence,homogenate extraction is a good alternative to extract polysaccharides from G.jasminoides.
