杜寒杂交羊Leptin基因外显子三的多态性及其与生长发育的相关性研究

摘　　要：利用引进的杜泊羊和本地的小尾寒羊杂交,培育地方优质肉羊新品种是山东省良种工程实施的任务之一。本研究利用DNA池和测序技术对杜寒杂交羊Leptin基因外显子三进行多态位点的筛查,再用PCR-RFLP技术对基因进行多态性分析,为在杂交后代中选育理想个体提供选择依据。研究结果表明,在试验群体202个样品中检测到G3941T一个突变位点G/T突变,将其基因型命名为GG、GT、TT,基因型频率分别为0.3020、0.5198、0.1782。统计分析表明,G3941T位点与生长发育性状存在相关性。在G3941T位点,TT基因型体重、体高、体长、胸围指标均高于GG基因型,其中体高和体长指标差异显著(P<0.05),体重和胸围指标差异极显著(P<0.01)。Leptin基因可以作为选育杜寒杂交羊生长发育性状的分子遗传标记。One of the major goals of Shandong Animal Breeding Project is to obtain superior meat sheep stocks by hybridizing the introduced Dopper Sheep with the local Small Tail Han sheep.This study aimed to examine the polymorphism of the exon 3 of Leptin gene by sequencing the DNA pools of the hybrids followed by PCR-RFLP,which may provide the basis for the selection of ideal individuals from the hybrids.In a test population of 202 individuals,a mutation of G/T was detected at the locus G3941T and the genotypes were thus named GG,GT,TT,with genotype frequencies of 0.3020,0.5198 and 0.1782,respectively.Statistical analyses showed that the genotype of G3941T correlated well with growth traits in that the phenotypes of the animals with TT genotype outperformed those with GG genotype in body weight （P ＜0.05）,body height （P ＜0.05）,body length （P ＜0.01） and chest circumference （P ＜0.01）.Leptin gene can be used as a molecular marker for breeding of Du Han hybrid sheep.
