摘  要:目的探讨脑源性神经营养因子(BDNF)对海湾战争综合征(GWS)海马细胞凋亡或存活的作用及其机制,为应用BDNF预防和/或治疗GWS提供理论依据。方法入选大鼠随机分为对照组、束缚应激组和GWS模型组。实验28 d结束后处死动物,取脑。采用免疫组化技术和免疫印迹分析检测海马CA1区BDNF及其受体酪氨酸蛋白激酶B(TrkB)和cAMP反应元件结合蛋白(CREB)的表达。结果束缚应激组大鼠海马BDNF、TrkB及CREB表达的平均吸光度值分别为0.291±0.001、0.253±0.002、0.215±0.013,明显低于对照组(BDNF 0.320±0.040、TrkB 0.243±0.004、CREB 0.232±0.009)(P<0.05);GWS模型大鼠海马BDNF、TrkB及CREB表达的平均吸光度值分别为0.269±0.036、0.226±0.004、0.194±0.010,明显低于低于对照组和束缚应激组(P<0.05)。结论 GWS模型大鼠海马CA1区BDNF及其受体TrkB以及调节因子CREB的表达明显下降。Objective To explore the role of hippocampal brain-derived neurotrophic factor(BDNF) in hippocampal cell apoptosis or survival in the Gulf War Syndrome (GWS) , so ao to provide theoretical evidence for using BDNF to prevent and/or treat GWS. Methods The rats were randomly divided into three groups: control group, restraint-stress group, and GWS group (established according to Abdel-Rahman et al). The animals were killed at the end of 28 d-experiment, and the brains were removed to detect the expression of the BDNF, tylrosine related receptor kinase B (TrkB) and cAMP response element binding protein (CREB) in the hippocampal CAI re- gion by using immunohistoehemical staining and Western blot analysis. Results The average absorbance of hipp- ocampal BDNF,TrkB ,and CREB in restraint-stress group (0.291 _±0. 001, 0. 253_±0.002, and 0.215_±0. 013 re- spectively) was significantly lower than those in control group (0.320±0. 040, 0. 243±0.004, 0. 232±0.009 ), (P〈0.05) ;the average absorbance of hippoeampal BDNF,TrkB, and CREB in GWS group (0.269±0. 036,0.226±0. 004, and 0. 194_±0. 010 respectively) was significantly lower than those in control group and restraint-stress group ( P〈0.05 ). Conclusion The expression of brain-derived neurotrophic factor, and its receptor( TrkB), as well as regulatory factor(CREB) in the hippocampal CA1 region of GWS rat model was significantly decreased.
