摘  要:目的:探讨PPARγ基因Pro12Ala多态性与妊娠期糖尿病之间的关系。方法:应用聚合酶链式反应-限制性内切酶技术,对104例妊娠期糖尿病(GDM)妇女和120例正常对照者PPARγ2基因Pro12Ala多态性位点进行基因分型。结果:PPARγ2Pro12Ala多态性的基因型频率PP、PA、AA,在GDM组和正常对照组中分别为92.4%、7.6%、0.0%以及85%、13.3%、1.7%。P等位基因和A等位基因在GDM组和对照组的频率分布分别为96.2%、91.7%和3.8%、8.3%。两组间的基因型和等位基因频率均无显著性差异(P>0.05)。在GDM组中,PPARγ2Pro12Ala多态性与体重指数呈正相关(P<0.01),而与空腹血糖、胰岛素、甘油三脂、总胆固醇无相关性。PPARγ2Pro12Ala多态性可能与GDM妇女肥胖的发生有一定的相关性,但与GDM无明显联系。Objective: To explore whether a peroxisome proliferator-activator receptor-γ2 (PPARγ2) Prol2Ala polymorphism was associated with gestational diabetes mellitus (GDM). Methods: In this case-control study, anthropometric and metabolic variables of 120 non-diabetic pregnant women and of 104 women who were diagnosed as GDM according to 75 g oral glucose tolerance test (OGTT) were compared on the basis of PPAR-y2 polymorphism by PCR-RFLP. Results: The genotype frequencies (PP,PA,AA) of the Prol2Ala variant in GDM and control group were 92.4%,7.6 % ,0.0% and 85 %,13.3%,1.7%. Frequencies of P allele and A allele were 96.2% vs 91.7% and 3.8% vs 8.3% in GDM group vs control group. There were no differences in geno- type and allele frequencie between these two groups(P〉0.05). But in the GDM group, there has a significant association between geneotype and the body mass index,while there was no differ- ence between geneotype and the FBS, FINS, TG and HOMA-IR. Conclusion: The PPAR-γ2 Prol2Ala polymorphism was not significantly associated with GDM, but it tends to increase the risk of obesity in GDM women.
